sB7H3在盲肠结扎穿孔脓毒症小鼠模型的表达特征及其意义

摘要 目的探索sB7H3在盲肠结扎穿孔(CLP)脓毒症小鼠血清表达特征及其意义。方法CLP法建立模型,50只健康昆明小鼠随机分为CLP组(术后12 h、术后24 h和术后48 h)、假手术组和对照组。CLP组、假手术组和对照组每组10只小鼠。术后12 h、24 h和48 h分别选取不同组别小鼠;先摘眼球取血,再收集腹腔灌洗液送培养。ELISA法检测血清C反应蛋白(CRP)、纤维蛋白原(FIB)和sB7H3水平。结果CLP组CRP和sB7H3均高于相应的对照组和假手术组(P<0.05),CLP组FIB均低于相应的对照组和假手术组(P<0.05)。CLP组内,术后48 h CRP均高于术后12 h和术后24 h(P<0.05),但术后12 h和术后24 h间CRP差异无统计学意义。CLP组内,术后48 h FIB低于术后12 h和术后24 h(P<0.05),但术后12 h和术后24 h FIB间差异无统计学意义。CLP组内,术后48 h sB7H3均高于术后12 h和术后24 h(P<0.05),术后24 h sB7H3也显著高于术后12 h(P<0.05)。CLP组腹腔灌洗液培养结果均为阳性,对照组和假手术组腹腔灌洗液培养结果均为阴性。结论盲肠结扎穿孔小鼠模型是研究脓毒症的可靠动物模型,sB7H3可能参与脓毒症进展。 Objective To explore the expression characteristics and significance of sB7H3 in serum of mice with sepsis induced by cecal ligation and perforation(CLP).Methods A mouse model of sepsis was established by CLP.Fifty healthy Kunming mice were randomly divided into CLP group(12 h,24 h and 48 h after operation),sham operation group and control group.Each group consisted of 10 mice.Mice from different groups were selected at 12 h,24 h and 48 h after operation,blood was extracted from eyeballs firstly and then peritoneal lavage fluid was collected for culture.The levels of C reactive protein(CRP),fibrinogen(FIB)and sB7H3 in the serum were detected by ELISA.Results The levels of CRP and sB7H3 in the CLP group were higher than those in the corresponding control and sham operation groups(P<0.05),and the FIB levels of CLP group were lower than those of the corresponding control group and sham operation group(P<0.05).In the CLP group,CRP levels at 48 h after operation were higher than those at 12 h and 24 h after operation(P<0.05),but there was no significant difference between CRP levels at 12 h and 24 h after operation.In the CLP group,FIB levels at 48 h after operation were lower than those at 12 h and 24 h after operation(P<0.05),but there was no significant difference between 12 h and 24 h.Within the CLP group,sB7H3 levels at 48 h after operation were higher than those at 12 h and 24 h after operation(P<0.05),and sB7H3 levels at 24 h after operation were significantly higher than those at 12 h after operation(P<0.05).The culture results of peritoneal lavage fluid in the CLP group were all positive,while those in the control and sham operation groups were all negative.Conclusion CLP mouse model is a reliable animal model for the study of sepsis and sB7H3 may be involved in the progression of sepsis.
