摘要
猪苓菌核际土壤微生物是构成猪苓菌核微生态系统的重要组成部分。为了深入研究长白山猪苓作为道地性药材的适生区,分析长白山区不同产地猪苓菌核际土壤的真菌多样性特征和群落组成,基于PacBio平台,利用单分子实时测序(SMRT Cell)方法对marker基因进行测序,测定长白山区不同产地猪苓菌核际土壤真菌群落结构。结果表明:所测样本共获得390 OTU,注释的真菌隶属于24纲53目97科168属。不同产地的猪苓菌核际土壤中基于门水平上子囊菌门是主要的优势菌门,基于属水平上被孢霉属为多数土壤样品的优势真菌属。土壤的理化因子对8个土样中真菌群落多样性的影响大小顺序为TP>TK>TN>pH。长白山区不同产地猪苓菌核际土壤的优势真菌群落功能存在明显差异,主要以外生菌根为主。
Intersoil microorganism is an important part of the nuclear microecosystem.In order to further study the suitable area of Polyporus umbellatus in Changbai mountain as authentic medicinal materials,the fungal diversity characteristics and community composition of the rhizosphere soil of Polyporus umbellatus from different areas in Changbai mountain were analyzed,and using PacBio platform real-time sequencing(SMRT Cell),the marker gene was sequenced to determine the structure of the soil in Changbai mountain.The results showed that a total of 390 OTU were obtained from the tested samples,and the annotated fungi belonged to 24 classes,53 orders,97 families and 168 genera.At the phylum level,Ascomycota is the main dominant fungal phylum in the rhizosphere soil of Polypo⁃rus umbellatus from different productions areas,while at the genus level,Aspergillus is the dominant fungal genus with most soil samples.The order of the effects of soil physicochemical factors on fungal community diversity in the eight soil samples is:TP>TK>TN>pH.The dominant fungi are obviously different in different production areas in Changbai mountain,mainly dominated by ectomycorrhizal fungi.
