参玫五味粉中总皂苷与多糖的提取及其体外活性研究

摘要 试验旨在探究参玫五味粉中总皂苷与多糖联合提取的最佳工艺条件及其体外活性。试验以参玫五味粉总皂苷与多糖的提取质量分数为考察指标,通过对提取溶剂浓度和浸泡时间的优化确定最佳提取工艺,测定参玫五味粉总皂苷提取物与多糖提取物对羟基自由基的清除率及对亚硝酸盐的抑制率,探索其体外抗氧化和抗肿瘤活性。结果显示:在最佳提取工艺条件下,联合提取4根层析柱中总皂苷的提取率依次为92.65%、98.99%、102.68%、104.31%,出膏率为10.00%,干浸膏中总皂苷含量为34.99 mg/g;多糖的提取率依次为93.57%、98.20%、100.77%、101.27%,出膏率为12.55%,干浸膏中多糖含量为19.20 mg/g。参玫五味粉总皂苷与多糖提取物清除羟基自由基的IC_(50) 值均大于对照VC,表明两者具有体外抗氧化活性,且具有一定的亚硝酸盐抑制作用。研究表明,柱层析联合提取法同时提取参玫五味粉总皂苷与多糖,提取率高且操作简便易行,可为畜牧业饲料添加剂的发展提供参考价值。 The experiment was to explore the optimal process conditions and invitro activity for the joint extraction of total saponins and polysaccharides from Shenmei Wuwei powder.The extraction mass fraction of total saponins and polysaccharides of Shenmei Wuwei powder was used as the investigation index by optimizing the extraction solvent concentration and soaking time,the optimal extraction process was determined,and the antioxidant and antitumor activities of Shenmei Wuwei Powder were determined by determining the scavenging rate of hydroxyl radicals and the inhibition rate of nitrite in total saponin extract and polysaccharide extract.The results showed that under optimal parameter conditions,the extraction rates of total saponins in the four chromatographic columns were 92.65%,98.99%,102.68%,and 104.31%,the paste yield was 10.00%,and the total saponin content was 34.99 mg/g.The extraction rate of polysaccharides was 93.57%,98.20%,100.77%and 101.27%,the ointment yield was 12.55%,and the polysaccharide content in dry extract was 19.20 mg/g.The IC_(50) values of saponin extract and polysaccharide extract for scavenging hydroxyl radicals were higher than those of control vitamin C,indicating that they had antioxidant activity invitro and certain nitrite inhibition.The study indicates that the total saponins and polysaccharides are extracted simultaneously by circulating column chromatography from the Shenmei Wuwei powder is high,and the operation is simple and easy,which provides reference value for the development of feed additives in animal husbandry.
