吉林黄鸡、北京油鸡及其杂交群体LEPR基因多态性与屠宰性能和肉质性状关联分析

摘要 为探讨地方鸡瘦素受体(LEPR)基因遗传多样性及其与生产性状的相关性,以吉林黄鸡、北京油鸡及其正反交杂群体为研究对象,通过基因组PCR直接测序挖掘LEPR基因多态位点,再采用竞争性等位基因特异性PCR(KASP)进行基因分型检测,最后采用单因素方差分析对各群体内不同基因型个体屠宰与肉质性状进行相关性分析。结果:LEPR基因内含子2存在3个SNP位点,其中g.735 G>A与报道一致;KASP法对g.427 C>T和g.735 G>A位点遗传多样性分析发现g.427 C>T位点在吉林黄鸡(HH)群体中T∶T为主要基因型,北京油鸡(YY)为C∶C型,g.735 G>A位点在4个群体中A∶G均为主要基因型,g.427 C>T位点除了HH群体外,其他群体均处于哈迪-温伯格不平衡状态,而g.735 G>A位点全都处于平衡状态;相关分析发现两位点均与腹脂率相关,但各群体间不同基因型并未表现明显的一致性,大部分群体中g.735 G>A位点不同基因型还与脚重,胸肌和腿肌滴水损失密切相关。综上,地方鸡LEPR基因多态位点及与屠宰和肉质性状相关分析为利用该基因进行优质鸡选育提供理论依据。 In this study,Jilin yellow chickens,Beijing oil chickens,and their ortho-and trans-breeding heterozygous populations were used as the research objects in order to investigate the genetic diversity of the LEPR gene in local chickens and the correlation of the diversity with the production traits of the chickens.The polymorphic sites of the LEPR gene were excavated by direct sequencing through genome PCR,followed by competitive allele specific PCR(KASP)for genotyping detection,and one-way ANOVA was used for the correlation analysis of slaughtering and meat quality traits of individuals of different genotypes in the different populations.Finally,a one-way analysis of variance(ANOVA)was employed to examine the relationship between the variables related to the meat quality and slaughter performance in the individuals with varying genotypes.The results were as follows:The genetic diversity analysis of the g.427 C>T and g.735 G>A loci by KASP showed that T∶T was the major genotype of g.427 C>T in Jilin yellow chickens(HH)and C∶C in Beijing oil chickens;and A∶G was the main genotype of g.735 G>A in all the four groups.Three SNPs loci were found in intron 2 of the LEPR gene,of which g.735 G>A was con sistent with the report.All the four populations had the predominant genotype A∶G,and all except the HH population had Hardy-Weinberg disequilibrium at the g.427 C>T locus,whereas all the populations showed equilibrium at the g.735 G>A locus;both the loci were linked to the percentage of abdominal fat,according to the correlation analysis.However,the various genotypes did not significantly differ between the populations;and in the majority of the populations,the various genotypes of the g.735 G>A locus were also linked to the foot weight and the loss of pectoral and leg muscle drip.To sum up,the examination of the polymorphic loci of the LEPR gene and its association with characteristics related to meat quality and slaughter in the local hens offered a theoretical foundation for the application of this gene in the breeding and selection of premium chickens.
