摘要
目的 建立HPLC法测定儿童维矿类口服溶液中苯甲酸和山梨酸含量的方法。方法 采用高效液相色谱法,二极管检测器,以C18色谱柱为分离柱,以甲醇-0.01 mol·L^(-1)乙酸铵溶液(用氨试液调节pH值至8.0,5∶95)为流动相,在230 nm处测定苯甲酸和山梨酸含量。流速为1.0 mL·min^(-1),进样体积为10μL。结果 苯甲酸和山梨酸分别在0.996 0~79.680 3μg·mL^(-1)和1.040 9~83.269 4μg·mL^(-1)范围内与峰面积呈良好的线性关系,平均回收率分别为97.86%、99.74%(n=9)。结论 本法操作快捷简便、结果可靠、重复性好,为儿童维矿类口服溶液中抑菌剂苯甲酸和山梨酸的质量控制提供了重要依据。
OBJECTIVE To establish an HPLC method for the determination of benzoic acid and sorbic acid in children's vitamin and mineral oral solutions.METHODS The HPLC method utilized a diode array detector and C18 column.The mobile phase was mixture of methanol and 0.01 mol·L^(-1) ammonium acetate solution(with the pH adjusted to 8.0 using ammonia solution,5∶95).The flow rate was 1.0 mL·min^(-1) and the detection wavelength was 230 nm.The injection volume was 10μL.RESULTS Benzoic acid and sorbic acid showed a good linear relationship with the peak area in the range of 0.9960 to 79.6803μg·mL^(-1) and 1.0409 to 83.2694μg·mL^(-1),respectively.The average recoveries were 97.86%(n=9)and 99.74%(n=9).CONCLUSION This method is accurate,reliable,and reproducible,and can be used for the determination of benzoic acid and sorbic acid in children's vitamin and mineral oral solutions.
