基于质谱的鳙鱼不同组织中内源性肽的分布研究

摘要 痕量内源性肽作为功能性生物活性物质广泛存在于生物体中,通常与细胞生长、物质代谢、免疫遗传等生理活动密切相关,并发挥着重要调节作用。由于不同组织与器官的生理功能不同,其中的内源性肽的种类和数量也存在差异。本文基于质谱技术对鳙鱼不同组织中的内源性肽加以研究,并探讨其种类与分布特征。基于磁固相微萃取(Magnetic Solid Phase Extraction,MSPE)技术,联合使用聚合物改性的磁性纳米材料Fe_(3)O_(4)@SiO_(2)@PAA和Fe_(3)O_(4)@PEI对样品中的内源性肽进行选择性富集,结合蛋白质组学方法对血液和不同脏器中的内源性肽进行差异比较和分析,并通过基因分析揭示其潜在的生理功能。结果表明,鱼脑中内源性肽的种类最多,而鱼肠中相对较少。质谱鉴定结果也说明鳙鱼体内不同组织与鱼血中的内源性肽存在很大差异,其种类与功能存在共性与个体差异,且差异性内源性肽与器官的生理结构和功能有所关联。这一研究结果初步揭示了鱼类不同组织中内源性肽的生理功能及生物过程的关系,为进一步进行空间蛋白质组学、不同组织的功能研究以及病毒入侵细胞的分子机制研究等提供了一定的理论依据。 Trace endogenous peptides exist widely in organisms as functional bioactive substances,which are usually closely related to physiological activities such as cell growth,substance metabolism,and immune genetics,and play an important regulatory role.Due to the different physiological functions of different tissues and organs,the types and quantities of endogenous peptides are obviously different.In this work,the endogenous peptides in different tissues of Bighead carp were studied by mass spectrometry,and their species and distribution characteristics were discussed.Based on magnetic solid phase extraction(MSPE),polymer-modified magnetic nanomaterials Fe_(3)O_(4)@SiO_(2)@PAA and Fe_(3)O_(4)@PEI were used for selective enrichment of endogenous peptides in samples.The differences of endogenous peptides in blood and different organs were compared and analyzed by proteomics method,and their potential physiological functions were revealed by gene analysis.The results showed that the number of endogenous peptides in fish brain was the highest,while the number in fish gut was the lowest.The results of mass spectrometry also showed that the endogenous peptides in different tissues and blood of bighead carp were quit different,and there were commonalities and individual differences in their types and functions,and the different endogenous peptides were related to the physiological structure and function of the organs.The results of this study initially revealed the relationship between the physiological functions and biological processes of endogenous peptides in different tissues of fish,and provided a theoretical basis for further studies on spatial proteomics,the function of different tissues and the molecular mechanism of virus invasion into cells.
