摘要
文章以产油酵母圆红冬孢酵母(Rhodosporidium toruloides)为宿主,测试来源于文冠果(Xanthoceras Sorbifolium)脂肪酸延长酶相关基因的异源表达。围绕文冠果的神经酸合成系统在R.toruloides中的重构可行性,调取文冠果的神经酸合成基因,并根据R.toruloides的密码子偏好性加以优化,构建表达载体。通过农杆菌介导将表达载体转化至R.toruloides单倍体菌株NP11中,并从表型、基因型及蛋白水平等方面入手验证脂肪酸延长酶FAE基因的表达。
The oil-producing yeast(Rhodosporidium toruloides)was the host to test the heterologous expression of fatty acid elongase related genes from the Xanthoceras Sorbifolium.According to the reconstruction feasibility of neural acid synthesis system of Xanthoceras Sorbifolium in R.toruloides,the neural acid synthesis gene of Xanthoceras Sorbifolium was obtained,and was optimized based on the codon bias of R.toruloides to construct the expression vector.And through Agrobacterium mediated,the expression vector was transformed into R.toruloides haploid strain NP11,and the expression of fatty acid elongase FAE gene was verified from the phenotype,genotype and protein level.
