摘要
E2F转录因子(E2F transcription factor,E2Fs)是视网膜母细胞瘤蛋白(Retinoblastoma protein,p Rb,RB或RB1)家族的主要靶蛋白.作为关键的转录因子家族成员,E2Fs在调控下游基因的转录、DNA合成和细胞周期中起到至关重要的作用.近年来,关于脂肪生成的研究显示,E2F家族的多个成员参与脂肪生成调控.综述了E2Fs在脂肪生成中的调控作用,主要包括E2Fs家族成员的结构特征及在脂肪生成中的作用机制.研究发现,E2F-1与E2F-3在哺乳动物中具有促进脂肪分化的功能,E2F-2可能参与牛前脂肪细胞分化过程,具体功能尚不清楚,而E2F-4,E2F-5是脂肪生成的抑制因子,在哺乳动物中起抑制脂肪生成作用,E2F-6,E2F-7,E2F-8在脂肪生成调控中的作用尚不明确.针对E2Fs在脂肪增殖和分化中的调控作用进行综述,探讨E2Fs作为肥胖靶点的治疗策略,为后续预防和治疗肥胖及相关疾病提供理论依据.
E2F transcription factors(E2Fs)are the main target proteins of the retinoblastoma protein(pRb,RB or RB1)family.As a key family member of the transcription factor,E2Fs play a crucial role in regulating the transcription,DNA synthesis and cell cycle of downstream genes.In recent years studies on adipogenic have shown that multiple members of the E2F family are involved in the regulation of adipogenic production.Reviewdthe regulatory role of E2Fs in adipogenesis,including the structural characteristics of E2Fs family members and the mechanism of action in adipogenesis.It was found that E2F-1 and E2F-3 had the function of promoting adipogenesis in mammals,and E2F-2 may be involved in the differentiation process of bovine preadipocytes,but the specific function is still unclear,while E2F-4 and E2F-5 are inhibitors of adipogenesis and play an inhibiting role in adipogenesis in mammals,while the role of E2F-6,E2F-7 and E2F-8 in the regulation of adipogenesis is not clear.Reviewd the regulatory role of E2Fs in fat proliferation and differentiation,and discusses the therapeutic strategy of E2Fs as a target for obesity,so as to provide a theoretical basis for the subsequent prevention and treatment of obesity and related diseases.
