The expression and prognostic significance of MED8 gene in glioma

目的通过CGGA和TCGA数据库,研究MED8基因在脑胶质瘤组织的表达与预后作用。方法通过TIMER2.0在线分析工具对TCGA数据库内胶质瘤及正常组织MED8基因表达差异进行研究,并进一步利用HPA数据库比较胶质瘤与正常组织中MED8蛋白的表达差异情况。通过CGGA数据库获取胶质瘤患者样本的mRNAseq_693和mRNAseq_325测序数据及其临床数据,利用R软件进行临床相关分析,预后生存分析以及单、多因素COX回归分析,同时绘制ROC曲线评价MED8在胶质瘤患者预后的作用;运用GSEA富集分析MED8在脑胶质瘤中的功能,然后进行基因共表达相关性分析。结果相较于正常组织,MED8在胶质瘤组织表达明显上调,同时,在胶质瘤组织中表达水平越高,胶质瘤患者预后越差,差异具有统计学意义(P<0.001)。此外,本研究发现MED8可作为独立生物标志物预测胶质瘤患者预后。结论在胶质瘤组织中MED8可能发挥促癌作用,MED8有望成为预测胶质瘤患者预后的新的生物标志物。

Objective To investigate the expression and prognostic significance of the MED8 gene in glioma tissues through analysis of the CGGA and TCGA databases.Methods Differential expression of the MED8 gene in gliomas compared to normal tissues was analyzed using the TIMER2.0 online tool for TCGA database and protein expression differences were assessed using the HPA database.Glioma patient samples from the CGGA database,including mRNAseq_693 and mRNAseq_325 sequencing data and clinical information,were analyzed using R software for clinical correlation analysis,prognostic survival analysis,and both univariate and multivariate COX regression analysis.Additionally,Receiver Operating Characteristic(ROC)curves were constructed to assess the prognostic significance of MED8 in the glioma patient cohort.Gene Set Enrichment Analysis(GSEA)was utilized to investigate the role of MED8 in gliomas,complemented by gene co-expression correlation analysis.Results MED8 expression was significantly elevated in glioma tissues compared to normal brain tissue,with higher expression levels associated with poorer prognosis in glioma patients(P<0.001).Furthermore,this study identified MED8 as an independent prognostic marker for predicting glioma patient outcomes.Conclusion The findings support the hypothesis that MED8 may play an oncogenic role in glioma tissues and highlight its potential as a novel biomarker for predicting the prognosis of glioma patients.
