摘要
糖基化修饰在生物体内扮演着重要的角色,广泛参与细胞黏附、分子运输、信号传导、受体激活等各种生理过程,同时也与癌症、感染性疾病、糖尿病等多种疾病的发生发展密切相关。糖基化是改善蛋白质和肽理化性质的有效策略,具体体现在稳定蛋白结构、赋予靶向性、提高半衰期等方面。合成均质糖蛋白和糖肽对研究基本生物学过程、疾病的诊断和治疗具有非常重要的意义。本文综述了糖基化对蛋白质和肽性质与功能的影响、自然界中存在的糖基化类型、糖基化修饰蛋白质和肽的方法以及部分已上市的糖蛋白和糖肽类药物,并对该领域的研究方向进行了展望。
Glycosylation modification plays an important role in organisms,widely participating in various physiological processes such as cell adhesion,molecular transport,signal transduction,receptor activation,etc.,and also closely related to the occurrence and development of cancer,infectious diseases,diabetes and other diseases. Glycation is an effective strategy to improve the physicochemical properties of proteins andpeptides, which is reflected in stabilizing protein structure, endowing targeting properties, and increasing halflife.The synthesis of homogeneous glycoproteins and glycopeptides is of great significance for studying basicbiological processes, diagnosing and treating diseases. This paper provides an overview of the effects ofglycosylation on the properties and functions of proteins and peptides, the types of glycosylation present innature, methods of glycopeptide modification of proteins and peptides, as well as some commercially availableglycoprotein and glycopeptide drugs. We also look forward to research directions in this field.
