大孔树脂分离纯化艾绒总黄酮工艺探索

摘要 探索优化大孔树脂分离纯化艾绒总黄酮的工艺。本实验采用单因素实验优化分离纯化工艺。经过研究,发现HPD100大孔树脂具有最优的吸附和解析附能力。采用90 min的上样时间,pH=3,吸附温度40℃,上样液质量浓度0.2 mg/mL,上样液流速1.5 m L/min,吸附率可达到68.5%。用体积分数80%乙醇洗脱,流速为1.5 m L/min,洗脱液用量为100 m L,洗脱率高达到60.45%。在此吸附和解析条件下,艾绒总黄酮的纯度由5.9%提升到13.5%,提纯效果好。 Explore and optimize the process of separating and purifying total flavonoids from Artemisia argyi using macroporous resin.This experiment uses a single factor experiment to optimize the separation and purification process.The results demonstrate that HPD100 macroporous resin has the most impressive adsorption and desorption capability.The ideal process conditions are:loading time of 90 minutes,pH of the solution at 3,adsorption temperature of 40℃,concentration of the solution at 0.2 mg/mL,flow rate of 1.5 mL/min,and adsorption rate of 68.5%.Elution was conducted with 80%ethanol by volume,with a flow rate of 1.5 mL/min and elution volume of 100 mL,resulting in a high elution rate of 60.45%.The purification effect was evidently effective,as the purity of total flavonoids in Artemisia argyi rose from 5.9%to 13.5%under these adsorption and desorption conditions.
