摘要
RNA剪接失调是肿瘤重要的分子病理特征之一,与剪接因子异常表达密切相关。多聚嘧啶串结合蛋白1(PTBP1)是剪接因子不均一核糖核蛋白家族成员之一,可通过调控增殖、迁移、侵袭、自噬、凋亡、免疫监视及代谢重编程等环节参与多种肿瘤的发生与发展,是消化道肿瘤的潜在临床应用靶点。PTBP1作为关键的剪接因子调节靶基因前体mRNA的剪接及mRNA稳定性等,其调控可变剪接的新型分子机制在消化道肿瘤中也得到了进一步阐述。该文对剪接因子PTBP1在消化道肿瘤中作用的研究进展作一综述。
Ribonucleic acid(RNA)splicing dysregulation is one of the important molecular pathological fea-tures of tumors,and RNA splicing dysregulation is closely related to abnormal expression of splicing factors.Polypyri-midine tract binding protein 1(PTBP1),a kind of RNA binding protein,is one of the members of the splicing factor heterogeneous nuclear ribonucleoproteins(hnRNPs)family,and is involved in the occurrence and progression of various tumors through regulation of proliferation,migration,invasion,autophagy,apoptosis,immune surveillance and meta-bolic reprogramming,and is a potential clinical application target for digestive tract tumors.PTBP1,as a key splicing factor,regulates the splicing of target gene precursor messenger RNA(mRNA)and the stability of mRNA,and its novel molecular mechanism for regulating alternative splicing has also been further elucidated in digestive tract tumors.In this paper,the research progress on the role of splicing factor PTBP1 in digestive tract tumors is reviewed.
