lncRNA-MHENCR对肾透明细胞癌细胞增殖、迁移和凋亡的影响

Effects of lncRNA-MHENCR on the proliferation,migration and apoptosis of clear cell renal cell carcinoma

目的 探讨黑色素瘤高表达竞争性长链非编码RNA(lncRNA-MHENCR)对肾透明细胞癌786-O细胞和Caki-1细胞增殖、迁移和凋亡的调控机制。方法 对癌症基因组图谱数据库中lncRNA-MHENCR在肾透明细胞癌的表达和预后进行分析;体外培养人肾透明细胞癌786-O和Caki-1细胞,分为对照组和si RNA组,培养72 h。采用CCK8法、克隆形成实验和EDU实验检测细胞增殖能力;Transwell小室实验检测细胞迁移能力;流式细胞实验和Western Blot检测肾透明细胞癌细胞凋亡情况。结果lncRNA-MHENCR与肾透明细胞癌的不良预后相关。敲低lncRNA-MHENCR抑制786-O和Caki-1细胞的增殖和迁移。流式细胞实验和Western Blot检测结果显示,敲低lncRNA-MHENCR表达可以使786-O和Caki-1细胞凋亡明显增加。Western Blot结果显示,敲低lncRNA-MHENCR表达使磷酸化蛋白激酶B和磷酸化哺乳动物雷帕霉素靶蛋白表达增加。结论 lncRNA-MHENCR可通过AKT/m TOR促进肾透明细胞癌增殖和迁移,并抑制肾透明细胞癌凋亡。

Objective To investigate the effects of the melanoma highly expressed competing endogenous long non-coding RNA(lncRNA-MHENCR)on the proliferation,migration,and apoptosis of clear cell renal cell carcinoma(ccRCC)786-O and Caki-1.Methods The expression and prognosis of lncRNA-MHENCR in TCGA were analyzed.The human ccRCC 786-O and Caki-1 were divided into control group and siRNA group and cultured for 72 h.The CCK8,colony formation assay,and EDU assay were utilized to detect cell proliferation.The transwell assay was used to detect cell migration.Flow cytometry and western blot were utilized to detect apoptosis.Results lncRNA-MHENCR was related to poor prognosis of ccRCC.lncRNA-MHENCR knockdown could inhibit proliferation and migration of 786-O and Caki-1.Results of flow cytometry and western blot confirmed that lncRNA-MHENCR knockdown could promote apoptosis of 786-O and Caki-1.The western blot showed that the expression of the phosphorylated protein kinase B(p-AKT)and phosphorylated mammalian target of rapamycin(p-mTOR)increased after lncRNA-MHENCR was knocked down.Conclusions lncRNA-MHENCR could promote proliferation and migration of ccRCC cells via AKT/mTOR,and inhibit apoptosis of ccRCC cells.
