miR-362及LncRNA SNHG12表达与IL-17甲基化与宫颈癌患者hr-HPV感染相关性

Association of expressions of miR-362,LncRNA SNHG12 and IL-17 methylation with hr-HPV infection in cervical cancer patients

摘要 目的探讨微小核糖核酸-362(miR-362)及长链非编码核糖核酸(RNA)小核仁RNA宿主基因12(Ln-cRNA SNHG12)表达水平、白细胞介素-17(IL-17)甲基化阳性率与宫颈癌(CC)患者高危型人乳头瘤病毒(hr-HPV)感染的相关性及生物学意义.方法选取2019年1月—2023年8月天津市第五中心医院收治的168例CC患者作为A组,同期收治的170例宫颈上皮内瘤变患者作为B组,180名同期健康体检女性志愿者作为C组;比较三组临床资料、hr-HPV感染率、宫颈组织miR-362、LncRNA SNHG12表达水平、IL-17甲基化阳性率,hr-HPV感染阳性、阴性宫颈组织miR-362及LncRNA SNHG12表达水平、IL-17甲基化阳性率,CC患者宫颈组织miR-362及LncRNA SNHG12表达水平、IL-17甲基化阳性率与hr-HPV感染阳性的相关性采用Spearman法进行分析,比较不同临床病理特征宫颈组织miR-362及LncRNA SNHG12表达水平,不同IL-17甲基化状态CC临床病理特征.结果A组宫颈组织miR-362表达水平低于B组、C组,B组低于C组;A组宫颈组织LncRNA SNHG12表达水平高于B组、C组,B组高于C组;三组IL-17甲基化阳性率分别为68.45%、52.35%、37.78%,差异有统计学意义(P<0.05);hr-HPV感染阳性CC患者宫颈组织miR-362水平低于hr-HPV感染阴性CC患者;宫颈组织LncRNA SNHG12表达水平高于hr-HPV感染阴性CC患者;IL-17甲基化阳性率为72.54%,高于hr-HPV感染阴性CC患者的46.15%(P<0.05);CC患者宫颈组织miR-362表达水平与hr-HPV感染阳性呈负相关(r=-0.565,P<0.05);宫颈组织LncRNA SNHG12表达水平、IL-17甲基化阳性率与hr-HPV感染阳性呈正相关(r=0.498、0.512,P<0.05);IL-17甲基化阳性患者肿瘤高分化程度的占比为60.87%,高于IL-17甲基化阴性患者的39.62%(P<0.05).结论宫颈组织miR-362、LncRNA SNHG12表达水平、IL-17甲基化阳性率与CC的发生及hr-HPV密切相关,且与患者临床病理特征存在一定关联. OBJECTIVE To explore the correlation and biological significance of the expression levels of microribo-nucleic acid-362(miR-362)and long-chain non coding ribonucleic acid(RNA)small nucleolar RNA host gene 12(LncRNA SNHG12),and the positive rate of interleukin-17(IL-17)methylation with high-risk human papilloma-virus(hr-HPV)infection in patients with cervical cancer.METHODS A total of 168 patients with cervical cancer admitted to the Fifth Central Hospital in Tianjin from Jan 2019 to Aug 2023 were enrolled and divided into the group A;170 patients with cervical intraepithelial neoplasia in the hospital were included in the group B,and 180 female volunteers under health examinations during the same period were in the group C.The clinical data and the occurrence rate of hr-HPV infection were compared among the three groups;the expression levels of cervical tis-sue miR-362 and LncRNA SNHG12 and positivity rate of IL-17 methylation among the three groups and between the hr-HPV-positive and negative cervical tissues were also compared.RESULTS The expression level of miR-362 in cervical tissue of the group A was lower than that of the group B and the group C,and the group B was lower than the group C.The expression level of LncRNA SNHG12 in cervical tissue of the group A was higher than that of the group B and the group C,and the group B was higher than the group C.The positive rates of IL-17 methyl-ation in the three groups were 68.45%,52.35%,and 37.78%,respectively,and the differences among them were statistically significant(P<0.05).The expression level of miR-362 in hr-HPV-positive CC patients was lower than that of HR-HPV-negative CC patients.The LncRNA expression level of SNHG12 in cervical tissues was higher than that of HR-HPV-negative CC patients.The positive rate of IL-17 methylation was 72.54%,which was higher than 46.15%in the hr-HPV-negative CC patients(P<0.05).The expression level of miR-362 in cervical tissue of cervical cancer patients was negatively correlated with hr-HPV-positive infection(r=-0.565,P<0.05),while the expression level of LncRNA SNHG12 and positivity rate of IL-17 methylation in cervical tis-sue were positively correlated with hr-HPV infection(r=0.498,0.512,P<0.05).The proportion of highly dif-ferentiated tumors in IL-17 methylation positive patients was 60.87%,higher than 39.62%in the IL-17 methyla-tion negative patients(P<0.05).CONCLUSION The expression levels of miR-362 and LncRNA SNHG12 in cer-vical tissue,as well as the positive rate of IL-17 methylation were closely related to the occurrence of cervical canc-er and hr-HPV infection,which has certain correlation with pathological characteristics of patients in clinic.
