不同透皮吸收促进剂及组合对烟酰胺凝胶体外透皮吸收的影响

摘要 目的研究不同透皮吸收促进剂及组合对烟酰胺凝胶体外透皮吸收效果的影响。方法采用Franz扩散池法,以小鼠腹部皮肤作为透皮屏障,用紫外-可见分光光度法测定接收液中烟酰胺的浓度,计算烟酰胺12h的累计渗透量(Q),考察不同的透皮吸收促进剂月桂氮?酮、卡必醇、二甲基亚砜、桉叶油、丁香油及它们组合使用的促渗透效果。结果每种含透皮吸收促进剂的烟酰胺凝胶的Q最高的分别是1%月桂氮卓酮(493.23±32.74)μg·cm^(-2)、1%卡必醇(607.14±37.79)μg·cm^(-2)、2%二甲基亚砜(657.6±39.37)μg·cm^(-2)、1%桉叶油(684.26±30.86)μg·cm^(-2)、1%丁香油(714.83±42.92)μg·cm^(-2),1%桉叶油+1%丁香油(689.38±34.45)μg·cm^(-2)。结论不论单一透皮吸收促进剂,还是透皮吸收促进剂组合均能促进烟酰胺的透皮渗透(P<0.05),其中1%丁香油对促进烟酰胺凝胶中烟酰胺的体外透皮吸收作用最佳。 Objective To study the effect of different transdermal absorption promoters and combinations on the transdermal absorption effect of nicotinamide gal.Methods Franz diffusion cell was used for in vitro osmotic experiment.The abdominal skin of mice was used as the test skin.The concentration of nicotinamide in the receiving solution was determined by UV spectrophotometry,and the cumulative release amount of nicotinamide(Q)was calculated.Research the effect of laurocapram,diethylene glycol monoethyl ether,DMSO,eucalyptus oil,clove volatile oil and combined transdermal absorption promoters..Results Q of nicotinamide gel with the best Single transdermal absorptions were 1%Laurocapram(493.23±32.74)μg·cm^(-2),1%Diethylene glycol monoethyl ether(607.14±37.79)μg·cm^(-2),2% DMSO(657.6±39.37)μg·cm^(-2),1% eucalyptus oil(684.26±30.86)μg·cm^(-2),1% clove volatile oil(714.83±42.92)μg·cm^(-2),1% eucalyptus oil+1% clove volatile oil(689.38±34.45)μg·cm^(-2).Conclusion Single and combined both have the effect of transdermal absorption,among all transdermal absorption promoters 1% clove volatile oil has the best effect.
