跨膜蛋白家族基因在肝癌患者中的预后模型构建评估

Construction and evaluation of prognostic model for transmembrane protein family genes in hepatocellular carcinoma

目的探讨跨膜蛋白(TMEM)家族基因对肝癌患者的预后价值,构建肝癌患者生存预后预测模型。方法筛选与肝癌患者预后相关的TMEM家族基因,采用LASSO回归模型构建预后预测模型,外部队列予以验证;基于风险评分和临床特征构建Nomogram图并验证其预测效能。结果纳入7个TMEM家族基因构建风险预测模型,低风险组肝癌患者预后显著优于高风险组。Cox回归分析结果显示该风险评分是肝癌患者预后的独立危险因素。基于风险评分构建的Nomogram图预测结果具有良好的契合度和应用价值。结论基于TMEM家族基因的预测模型可以很好地预测肝癌患者预后,为临床早期识别预后不良的肝癌患者提供一定理论依据。

Objective To explore the mechanisms and prognostic value of the Transmembrane protein(TMEM)family genes in liver cancer,and construct survival and prognosis prediction models for hepatocellular carcinoma(HCC).Methods TMEM family genes associated with the prognosis of HCC were analyzed and identified.The LASSO regression model was used to construct the prognostic prediction model,which was validated by an external cohort.The Nomogram was constructed based on the risk scores and clinical characteristics,and its predictive efficacy was verified.Results Seven TMEM family genes were included to construct the risk prediction model.HCC patients in the low-risk group had significantly better outcomes than those in the high-risk group.The COX regression analysis showed that the risk score was an independent risk factor for the prognosis of liver cancer patients.In addition,Nomogram′s prediction results based on risk scores were well-fitting and useful for applications.Conclusion The prediction model based on the TMEM family genes can effectively predict the prognosis of HCC and offer a theoretical basis for the early identification of patients with poor prognosis of HCC.
