鹿源多杀性巴氏杆菌辽宁株的分离鉴定及耐药性分析

摘要 研究旨在确证鹿源多杀性巴氏杆菌的血清型和耐药性等特征,无菌采集某养鹿场发病鹿的心脏、肝脏、肺脏、脾脏等病料,进行细菌分离培养、分子生物学鉴定、生化试验及致病性试验、药敏试验。结果显示,通过细菌学和分子生物学方法进行鉴定,确认分离到一株鹿源多杀性巴氏杆菌,分离株在10%绵羊鲜血营养琼脂培养基上形成圆形光滑、半透明的菌落,瑞氏染色呈两极浓染的小短杆状菌;通过血清型特异引物鉴定方法确认分离株为荚膜血清型D型。致病性试验结果显示,小鼠感染分离株后发病率为100%,死亡率为50%,并表现肾脏、肝脏及肠道黏膜等多器官出血和变质性变化。分离株对氨苄西林、阿莫西林/克拉维酸、美罗培南等12种药物敏感。研究表明,试验分离的鹿源多杀性巴氏杆菌具有较强的致病性,且对大多数抗菌药物仍具有敏感性,可能与鹿的生存环境和日常很少用药有关。 The study aimed to confirm the serotype and drug resistance characteristics of Pasteurella multocida from deer source.Sterile collection of diseased tissues such as heart,liver,lung,and spleen from the sick deer in a deer farm was conducted for bacterial isolation,culture,molecular biological identification,biochemical tests,pathogenicity tests,and drug sensitivity tests.The results showed that one strain of Pasteurella multocida from deer source was identified through bacteriological and molecular biological methods.The isolated strain formed round,smooth,translucent colonies on 10%sheep blood agar medium,and Wright's staining revealed small coccobacilli with polar intensification.The serotype-specific primer identification method confirmed the strain as capsular serotype D.Pathogenicity tests indicated that the infection rate of mice after infection with the isolated strain was 100%,with a mortality rate of 50%,and exhibited multiple organ hemorrhages and degenerative changes in the kidneys,liver,and intestinal mucosa.The isolates were sensitive to 12 drugs,including ampicillin,amoxicillin/clavulanic acid and meropenem.The study indicated that the Pasteurella multocida from deer source had strong pathogenicity and was still sensitive to most antibiotics,which may be related to the living environment of deer and few daily drugs.
