石莼酶解提取物延缓皮肤衰老作用

摘要 目的:研究石莼酶解提取物对D-半乳糖致衰老模型小鼠的抗衰老作用。方法:按800 mg/kg·d进行颈背部皮下注射D-半乳糖6周,建立小鼠亚急性衰老模型;提取物和V_(C)给药28 d后,表征小鼠皮肤老化及氧化应激水平。结果:与水浸提物相比,石莼酶解提取物中可溶性总糖、蛋白质、还原糖、多酚固形物含量显著提高(P<0.05),且对羟自由基和DPPH自由基清除率及SOD活力提高5.72、7.95、13.26倍。动物实验显示,石莼酶解提取物显著改善衰老性皮肤GSH-Px、T-SOD、CAT、T-AOC活力及MDA、蛋白质羰基水平,缓解小鼠脏器萎缩;皮肤中胶原蛋白含量提高71.63%,Ⅰ/Ⅲ型胶原比例提高55.40%;显著提高机体抗氧化酶GSH-Px、T-SOD、CAT活力和T-AOC水平;抑制机体生物大分子的氧化降解,如降低蛋白质羰基和MDA水平(P<0.05)。结论:石莼酶解提取物可降低小鼠氧化应激水平,延缓小鼠皮肤衰老。 Objective:The anti-aging effect of enzymatic hydrolysate extract of Ulva lactuca L.on D-galactose-induced aging model mice was studied.Methods:D-galactose was subcutaneously injected into the neck and back of mice at a dose of 800 mg/kg·d for 6 weeks to establish a subacute aging model.After 28 days of administration of the extract and Vitamin C,the skin aging and oxidative stress levels of the mice were characterized.Results:Compared with the water extract,the enzymatic hydrolysate extract of Ulva lactuca L.showed significantly higher levels of soluble total sugars,proteins,reducing sugars,and polyphenol solids(P<0.05),and increased the clearance rates of hydroxyl and DPPH free radicals and superoxide dismutase(SOD)activity by 5.72,7.95,and 13.26 times,respectively.Animal experiments showed that the enzymatic hydrolysate extract of Ulva lactuca L.significantly improved the glutathione peroxidase(GSH-Px),total superoxide dismutase(T-SOD),catalase(CAT),total antioxidant capacity(T-AOC),and malondialdehyde(MDA)and protein carbonyl levels in aging skin,and alleviated organ atrophy in mice.The collagen content in the skin increased by 71.63%,and the ratio of type Ⅰ/Ⅲ collagen increased by 55.40%.The enzymatic hydrolysate extract of Ulva lactuca L.significantly increased the GSH-Px,T-SOD,and CAT activity and T-AOC level in the body,and inhibited the oxidative degradation of biomolecules in the body,such as reducing protein carbonyl and MDA levels(P<0.05).Conclusion:The enzymatic hydrolysate extract of Ulva lactuca L.can reduce oxidative stress levels in mice and delay skin aging.
