摘要
采用超声辅助低共熔溶剂法制备蛹虫草多糖。以多糖得率为指标,先从4种常用的低共熔溶剂体系中确定最佳溶剂,然后通过单因素试验和响应面试验获得最佳提取工艺,并对其抗氧化活性进行评估。结果表明:最佳低共熔溶剂为氯化胆碱-尿素、摩尔比为1∶2、含量为30%;最佳提取工艺为液料比53∶1(mL/g)、超声温度51℃、超声时间37 min。在此条件下,多糖得率为21.56%±0.45%。抗氧化试验结果表明,蛹虫草多糖对DPPH自由基、羟基自由基和ABTS自由基的EC_(50)分别为0.80、1.38、0.36 mg/mL,表明其具有较好的抗氧化活性。
Cordyceps militaris polysaccharides was prepared by ultrasonic-assisted deep eutectic solvent method.Taking polysaccharide yield as an indicator,the optimal solvent was first determined from four commonly used deep eutectic solvent systems.Then,the best extraction process was obtained through single factor experiments and response surface experiments,and its antioxidant activity was evaluated.The results showed that the optimal deep eutectic solvent was choline chloride-urea,with molar ratio of 1∶2 and content of 30%.The optimal extraction process was liquid to material ratio 53∶1(mL/g),ultrasonic temperature 51℃,and ultrasonic time 37 min.Under these conditions,the polysaccharide yield was 21.56%±0.45%.The antioxidant test results showed that the EC_(50)of Cordyceps militaris polysaccharides on DPPH free radicals,hydroxyl free radicals,and ABTS free radicals were 0.80,1.38,and 0.36 mg/mL,respectively,indicating that it had good antioxidant activity.
