超声波辅助酶法提取向日葵花盘总黄酮及其抗氧化活性研究

摘要 以向日葵花盘为原料,采用超声波辅助酶法提取总黄酮,以其提取量为指标,在单因素试验的基础上采用响应面法优化提取工艺,并研究向日葵花盘总黄酮的抗氧化活性。结果表明:最佳提取工艺条件为酶解时间1 h、纤维素酶添加量0.1%(以向日葵花盘质量计)、超声时间24 min、超声功率194 W,在此条件下总黄酮提取量可达(20.45±0.13)mg/g。向日葵花盘总黄酮对DPPH自由基清除率和ABTS^(+)自由基清除率的IC_(50)分别为0.131、0.441 mg/mL,表明向日葵花盘总黄酮具有较强的抗氧化活性。 Using sunflower disks as raw materials,total flavonoids were extracted using ultrasound assisted enzymatic method.The extraction amount was used as the indicator,and the extraction process was optimized by response surface methodology based on single factor experiments.The antioxidant activity of total flavonoids in sunflower disks was studied.The results showed that the optimal extraction process conditions were enzymatic hydrolysis time 1 h,cellulase addition 0.1%(based on the mass of sunflower disks),ultrasonic time 24 min,and ultrasonic power 194 W.Under the conditions,the total flavonoids extraction amount could reach(20.45±0.13)mg/g.The IC_(50)of total flavonoids in sunflower disks for DPPH radical scavenging rate and ABTS^(+)radical scavenging rate were 0.131,0.441 mg/mL,respectively,indicating that total flavonoids in sunflower disks had strong antioxidant activity.
