PYNOD抑制NLRP3炎症小体调控小胶质细胞焦亡的脓毒症相关性脑病

PYNOD regulates microglial pyroptosis by inhibiting NLRP3 inflammasome in sepsis-associated encephalopathy

摘要 目的观察PYNOD对小胶质细胞焦亡的影响。方法采用盲肠结扎穿刺构建脓毒症小鼠模型,收集小鼠脑组织,采用蛋白质免疫印迹(Western blot)检测小鼠脑组织中PYNOD水平。通过LPS刺激BV2细胞或过表达PYNOD,采用蛋白质免疫印迹检测细胞中PYNOD与GSDMD蛋白表达水平。结果与假手术组相比,脓毒症组小鼠脑组织中PYNOD表达水平显著增加;与对照组相比,LPS刺激后BV2细胞中PYNOD表达显著增加;PYNOD高表达能够显著抑制GSDMD水平。结论脓毒症模型小鼠脑组织中PYNOD表达水平升高,下调GSDMD表达,抑制小胶质细胞焦亡。 Objective To investigate the effects PYNOD exerted on microglial cells.Methods Cecal ligation puncture(CLP)was utilized to establish a sepsis animal model and western blot was used to detect the expression of PYNOD in brain tissue.Besides,BV2 cells were transfected with PYNOD overpressing plasmid or LPS stimulation.Then,western blot was utilized to investigate the expression of PYNOD and GSDMD.Results PYNOD was significantly increased in the sepsis group,compared to the sham group,which is consistent with the cellular level in BV2 cells.Besides,PYNOD overexpression acted to inhibit the expression of GSDMD.Conclusions High expression of PYNOD in brain tissue of sepsis mice acted to down-regulation of GSDMD expression,suppressing microglial cell pyroptosis.
