摘要
以绿原酸(CA)为碳源,通过溶剂热法一步合成绿原酸衍生的碳基发光材料CA-71。借助荧光光谱、细胞共聚焦成像及细胞活力实验,进行CA-71荧光性质及细胞毒性研究。结果表明,绿原酸碳基材料CA-71荧光发射光谱具有激发依赖特性,最大激发波长为400 nm,对应的发射波长为492 nm。共聚焦成像实验进一步显示,CA-71能够通透细胞膜,定位人脐静脉内皮细胞(HUVEC)内线粒体,与线粒体绿色定位染料高度定位重合,皮尔森系数为0.98。此外,25.00 mg/L的CA-71对体外HUVEC细胞24 h抑制率为(7.70±0.93)%,与等质量浓度绿原酸对HUVEC细胞的抑制率(8.24±1.18)%接近,总体呈现低毒性,但无显著性差异。
Chlorogenic acid(CA)-derived luminescent carbon material CA-71 was synthesized by one-step solvothermal method.Fluorescence spectroscopy,confocal imaging and in vitro cell viability experiments were conducted to investigate the fluorescence properties and cytotoxicity of CA-71.It was indicated that the fluorescence emission spectrum of CA-71 exhibited excitation-dependence,giving the maximum excitation wavelength at 400 nm and the corresponding emission 492 nm.In confocal imaging experiments on human umbilical vein endothelial cells(HUVEC)in vitro,CA-71 showed membrane permeability and could target mitochondria in HUVEC cells with a high-degree overlap of CA-71 and Mito-Tracker Green,giving a pearson's coefficient 0.98.In addition,the 24 hour-inhibition of CA-71 at 25.00 mg/L on HUVEC cells in vitro was(7.70±0.93)%,which was close to the inhibitory effect(8.24±1.18)%for chlorogenic acid,showing low-cytotoxicity and no significant difference.
