共载黄芪甲苷-黄连素/白及多糖微球的制备与性能研究

摘要 利用白及多糖的生物黏附性制备共载黄芪甲苷-黄连素/白及多糖微球(AS-IV-BBR/BSP-Ms),并对其性能进行评价。结果表明,采用乳化交联法制备得到共载AS-IV-BBR/BSP-Ms,微球粒径在30~40μm,Zeta电位为-21.4 mV,且30 min内溶胀率达54.3%;红外光谱分析、X-射线衍射分析表明,黄芪甲苷和黄连素被包裹于白及多糖微球内部;DSC分析表明,在t=121℃时,出现尖锐吸热峰,热稳定性较好;体外抗氧化活性实验表明,该微球DPPH自由基的半数清除浓度(IC_(50))为8.23 mg/mL。该制备工艺稳定可行,微球性能评价达到预期目标,可为下一步药效学评价提供实验依据。 To prepare co-Delivery Astragaloside IV-berberine with Bletilla striata polysaccharide Microspheres(Ms)was prepared by using the bioadhesive property of bletilla striata polysaccharide and evaluate their properties were evaluated.The results showed that co-delivery Astragaloside IV-berberine with bletilla striata polysaccharide microspheres(AS-IV-BBR/BSP-Ms)were prepared by emulsion crosslinking method,the particle size of the drug-carrying microspheres was 30~40μm,the zeta potential was-21.4 mV,the swelling ratio could reach upwards of 54.3%in 30 min.The infrared spectroscopy and X-ray diffraction analysis showed that astragaloside IV and berberine were wrapped in the white and polysaccharide microspheres.DSC analysis showed that sharp endothermic peak appeared at about 121℃with good thermal stability.The in vitro antioxidant activity experiments showed that the DPPH radical scavenging IC_(50)Was 8.23 mg/mL.The preparation process was stable and feasible,and the properties evaluation of microspheres reached the expected goal,which could provide experimental basis for the next pharmacodynamic evaluation.
