针刺对PSCI模型大鼠海马蛋白BDNF、TrKb表达的影响

Effect of acupuncture on the expression of brain-derived neurotrophic factor and tyrosine kinase receptors B proteins in the hippocampus of rat model of post-stroke cognitive impairment

摘要 目的观察针刺对卒中后认知障碍(PSCI)模型大鼠海马脑源性神经营养因子(BDNF)/酪氨酸激酶受体B(TrKb)信号通路的影响,探究针刺疗法对PSCI的改善作用及其可能作用机制。方法40只SD雄性大鼠,随机取8只为假手术组(Sham),其余大鼠采用线栓法建立右侧大脑中脑动脉闭塞(MCAO)模型,造模完成后进行Zea-Longa神经功能评分和Morris水迷宫筛选认知障碍大鼠,并随机分为模型组(MCAO)、针刺组(MAS)。MAS组给予百会、神庭、风府、神门针刺治疗,每日1次,每次15 min,每周6次,共28 d。Sham组及MCAO组仅进行抓摸后放回笼中。结果Zea-Longa神经功能评分和水迷宫实验结果显示,MAS组较MCAO组神经缺损和学习记忆能力改善更明显(P<0.05);HE染色结果显示,MAS组较MCAO组神经元变性减少,组织结构紧凑,神经元数量增多;WB、RT-PCR结果显示,MAS组较MCAO组BDNF、TrKb表达水平上升(P<0.05)。结论针刺可提高PSCI模型大鼠学习记忆能力,其机制可能与促进BDNF/TrKb信号通路有关。 Objective To observe the effect of acupuncture on the brain-derived neurotrophic factor(BDNF)/tyrosine kinase receptors B(TrKb)signaling pathway in the hippocampus of rat model of post-stroke cognitive impairment(PSCI),and to explore the improvement effect of acupuncture therapy on PSCI and its possible mechanism of action.Methods 8 SD male rats were randomly selected as the sham-operated group(Sham group)from 40 SD male rats,while the remaining rats were used to establish the model of right middle cerebral artery occlusion(MCAO)using suture-occluded method.After modeling,the neurological function status of rats was assessed using the Zea-Longa score,and cognitive impairment rats were screened using Morris water maze.And then successfully modeled ras were randomly divided into the model group(MCAO group)and the acupuncture group(MAS group).The MAS group received acupuncture treatment at Baihui,Shenting,Fengfu,and Shenmen points,once a day,15 minutes each time,6 times per week,for a total of 28 days.The Sham group and MCAO group were only touched by hands,and then were placed back in their cages.Results The results of Zea-Longa score and Morris water maze test showed that the improvement in neurological deficits and learning and memory abilities in the MAS group was more significant than that in the MCAO group(P<0.05);HE staining results showed that compared with the MCAO group,the MAS group had reduced degeneration of neurons,compact tissue structure,and increased number of neurons.The results of RT-PCR showed that compared with the MCAO group,the expression levels of BDNF and TrKb in the MAS group were increased(P<0.05).Conclusion Acupuncture can improve the learning and memory abilities of PSCI model rats,and its mechanism may be related to the promotion of the BDNF/TrKb signaling pathway.
