17种中草药体外抗炎作用研究

摘要 采用超声提取法利用水和乙醇分别获得17种中草药的水和醇提物,将其作用于脂多糖(LPS)诱导的RAW264.7细胞模型,通过检测提取物对细胞存活率和炎症介质一氧化氮(NO)、白介素-6(IL-6)、白介素-1β(1L-1β)、肿瘤坏死因子-α(TNF-α)的影响,综合评价其抗炎效果,并研究其作用机制。结果表明:黄花蒿、冷蒿、白头翁、龙蒿、白鲜果皮、多叶棘豆、斜茎黄耆种皮、灵芝菌棒、苍术根须、山韭醇提物和黄花蒿、冷蒿、广布野豌豆、东北珍珠梅、苍耳、茜草水提物无细胞毒性;通过评估无细胞毒性醇和水提物抗炎活性,最终证明冷蒿水提物具有更好的抗炎作用,可显著降低IKKβ、p65和IKKα表达量,揭示了其可通过调控NF-κB通路发挥抗炎作用。本试验结果为冷蒿防治畜禽细菌性肠炎的研发及应用提供理论依据。 Using the ultrasonic extraction method and distilled water and ethanol as extraction solvents,aqueous extract and alcohol extract of 17 different Chinese herbal medicines were obtained.It was then applied to the LPS-induced RAW264.7 cells model to assess the anti-inflammatory effect by detecting the effects of the extract on cell survival rate and inflammatory mediators(NO,IL-6,11L-1β,TNF-α),to identify the extract with the best anti-inflammatory effect,and to investigate its mechanism of action.The result showed that the ethanolic extracts of Artemisia annua,Artemisia frigida Wild.,Pulsatilla chinensis,Artemisia dracunculus L.,pericarps of Dictamnus dasycarpus Turcz.,Oxytropis polyphylla DC.,seed coat of Astragalus adsurgens Pall.,fungus rods of Ganoderma lucidum,Atractylodes Lancea DC.,Allium senescens L.and the aqueous extract of Artemisia annua,Artemisia frigida Wild.,Vicia cracca L.,Sorbaria sorbifolia L.,Xanthium sibiricum L.,Rubia cordifolia L.had no cytotoxic effect on the cells.By evaluating the in vitro anti-inflammatory activity of ethanolic extract and aqueous extract of Chinese herbal medicine,which revealed that the aqueous extract of Artemisia frigida Wild.had the best anti-inflammatory capacity which may exert anti-inflammatory effects by inhibiting the mRNA expression of IKKβ,p65 and IKKαin the NF-κB signaling pathway.These results provide a theoretical basis for the development and application of Artemisia frigida Wild.for the control of bacterial enteritis in livestock and poultry.
