黄花倒水莲总黄酮提取工艺优化与抗氧化活性研究

摘要 近年来,临床生产中越来越多的黄酮类活性成分被添加到畜禽饲料中,用于改善产品质量、预防和治疗疾病等,具有广泛的应用前景。探索黄花倒水莲中总黄酮的提取工艺,并将其应用于临床生产是一项非常有前景和意义的研究。本试验基于单因素筛选和响应面优化法,以黄花倒水莲总黄酮提取率为指标,使用超声微波法从黄花倒水莲中提取总黄酮。通过对料液比、乙醇浓度、超声功率、提取时间和超声温度进行单因素试验研究,基于单因素结果差异,选取提取时间、乙醇浓度、料液比3个因素进行响应面工艺优化,确定黄花倒水莲总黄酮的最佳提取工艺,并制得黄花倒水莲总黄酮提取物。此外,采用DPPH·、ABTS+·自由基清除试验对黄花倒水莲总黄酮进行体外抗氧化活性研究。结果表明,黄花倒水莲总黄酮最优提取工艺条件为料液比1∶17(g∶mL)、乙醇浓度70%、提取时间60 min、超声功率225 W、超声温度60℃,最终得到黄花倒水莲总黄酮的提取率为3.63%,与软件预测最优工艺提取率相近,表明该工艺稳定、可行;质量浓度为1.00 g/L的黄花倒水莲总黄酮提取物对DPPH·、ABTS+·自由基清除率最高,分别为72.13%、68.13%,表明黄花倒水莲总黄酮提取物具有良好的体外抗氧化活性。本研究确定了黄花倒水莲总黄酮的最佳提取工艺,并证明其在体外具有较强的抗氧化活性,为黄花倒水莲总黄酮的进一步开发和利用奠定了理论基础。 In recent years,more and more flavonoid active ingredients have been added to livestock and poultry feeds in clinical production for improving product quality,preventing and treating diseases,etc.,which has a wide range of application prospects.Exploring the extraction process of total flavonoids from Polygala fallax Hemsl.and applying them to clinical production is a very promising and meaningful research.In this experiment,total flavonoids were extracted from Polygala fallax Hemsl.by ultrasonic microwave method based on one-way screening and response surface optimization method by the extraction rate of total flavonoids from Polygala fallax Hemsl.as an indicator.Single-factor comparative test was conducted to investigate the material-liquid ratio,ethanol concentration,ultrasonic power,ultrasonic time and ultrasonic temperature;based on the difference of single-factor results,three factors,namely,ultrasonic time,ethanol concentration and material-liquid ratio were selected for the response surface process optimization,to determine the optimal extraction process of Polygala fallax Hemsl.total flavonoids and obtain the Polygala fallax Hemsl.total flavonoid extracts;moreover,the in vitro antioxidant activity of Polygala fallax Hemsl.total flavonoid extracts was investigated using DPPH·,ABTS+·free radical scavenging assay.The results showed that the optimal extraction process conditions for Polygala fallax Hemsl.total flavonoids were material-liquid ratio of 1∶17(g∶mL),ethanol concentration of 70%,extraction time of 60 min,ultrasonic power of 225 W,and ultrasonic temperature of 60℃,the final extraction rate of Polygala fallax Hemsl.total flavonoids was 3.63%,which was similar to the software prediction of the optimal process,indicating that the process was stable and feasible.The in vitro antioxidant results showed that Polygala fallax Hemsl.total flavonoids at a concentration of 1.00 g/L had the highest scavenging rate of DPPH·and ABTS+·free radicals,which were 72.13%and 68.13%,respectively,indicating that Polygala fallax Hemsl.total flavonoids had good antioxidant activity in vitro.In conclusion,the optimal extraction process of Polygala fallax Hemsl.total flavonoids is obtained and the total flavonoids has good antioxidant activity in vitro,which lays a theoretical foundation for the further development and utilization of Polygala fallax Hemsl.total flavonoids.
