响应面法优化沙枣叶多酚提取工艺及体内外抗氧化活性研究

Optimization in Extraction Process of Polyphenols from Elaeagnus angustifolia L.Leaves by Response Surface Methodology and Its Antioxidant Activities in vitro and in vivo

摘要 采用单因素实验和响应面法优化了新疆沙枣叶多酚的超声辅助提取工艺,并通过测定沙枣叶多酚对DPPH自由基的清除能力及对小鼠血清中丙二醛(MDA)、超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)、总抗氧化能力(T-AOC)的影响评价其抗氧化活性。结果表明,沙枣叶多酚的最佳提取工艺为:料液比1∶40(g∶mL)、乙醇体积分数58%、提取时间21 min、提取功率250 W,在此条件下,多酚含量为36.45 mg·g^(-1);当浓度为0.20 mg·mL^(-1)时,沙枣叶多酚和VC对DPPH自由基的清除率分别为90.3%、96.5%;与空白对照组比较,模型对照组小鼠血清中MDA水平极显著升高(P<0.01),SOD、GSH-Px、T-AOC的水平均极显著降低(P<0.01),且模型对照组的肾脏系数和肝脏系数与空白对照组差异极显著(P<0.01);与模型对照组比较,沙枣叶多酚高剂量组血清中MDA水平极显著降低(P<0.01),SOD、GSH-Px、T-AOC的水平均极显著升高(P<0.01),且肾脏系数和肝脏系数与模型对照组无显著差异(P>0.05)。 We optimized the ultrasonic-assisted extraction process of polyphenols from Xinjiang Elaeagnus angustifolia L.leaves by single-factor experiments and response surface methodologies.Moreover,we evaluated the antioxidant activity of polyphenols from Elaeagnus angustifolia L.leaves through the determination of scavenging ability of polyphenols to DPPH free radicals and the effects of polyphenols on the expression levels of malondialdehyde(MDA),superoxide dismutase(SOD),glutathione peroxidase(GSH-Px),and total antioxidant capacity(T-AOC)in serum of mice.The results show that the optimal extraction process is determined as follows:the solid-liquid ratio of 1∶40(g∶mL),the ethanol volume fraction of 58%,the extraction time of 21 min,and the extraction power of 250 W.Under above conditions,the polyphenol content can reach 36.45 mg·g^(-1).When the concentration is 0.20 mg·mL^(-1),the scavenging rates of DPPH free radicals by polyphenols and VC are 90.3%and 96.5%,respectively.Compared with blank control group,MDA level in serum of model control group increases extremely significantly(P<0.01),the levels of SOD,GSH-Px,and T-AOC decrease extremely significantly(P<0.01),and renal coefficient and liver coefficient of model control group are extremely significantly different from those of blank control group(P<0.01).Compared with model control group,MDA level in serum of high-dose polyphenol group decreases extremely significantly(P<0.01),the levels of SOD,GSH-Px,and T-AOC increase extremely significantly(P<0.01),and renal coefficient and liver coefficient are not significantly different from those of model control group(P>0.05).
