摘要
病毒RNA的检测需要阳性对照或可定量的标准品。人们起初使用灭活或减毒的病原体和体外转录RNA作为阳性对照用于核酸检测,但使用病原体存在生物安全风险,且RNA容易降解及无法对RNA病毒从核酸提取到检测全过程监测,因此一种新的技术,即“装甲RNA技术”被用于高传染性及致病性RNA病毒的检测。噬菌体MS2是广泛存在于自然界中的一种单链RNA病毒,可利用噬菌体MS2的衣壳蛋白对目的RNA进行封装和保护,提高RNA的稳定性。本文针对基于噬菌体MS2的装甲RNA技术及其在病毒核酸检测中的应用进行文献综述。
The detection of viral RNA requires positive control or quantifiable standard.Initially,inactivated or attenuated pathogens and transcribed RNA in vitro were used for nucleic acid detection as positive controls.However,the use of pathogens poses biosafety risks,and RNA is prone to degradation and cannot be monitored throughout the entire process from nucleic acid extraction to detection.Therefore,a new technology,the"armored RNA technology",has been used for the detection of highly infectious and pathogenic RNA viruses.Bacteriophage MS2 is a single-stranded RNA virus that widely exists in nature.Armored RNA technology refers to the use of the capsid protein of bacteriophage MS2 to encapsulate and protect the target RNA to improve the stability of the RNA.In this paper,the phage MS2-based armored RNA technology and its application in viral nucleic acid detection are reviewed.
