响应面法优化菊苣叶黄酮提取工艺及其抗氧化活性分析

摘要 试验旨在提高菊苣叶的饲用价值。试验通过响应面法优化菊苣叶中黄酮的提取工艺,评估其抗氧化活性。在单因素试验的基础上,以黄酮得率作为响应变量,对影响菊苣叶中黄酮得率的3个因素进行Box-Behnken设计与优化,分析提取过程中液料比、提取温度和提取时间对菊苣叶黄酮得率的影响。结果表明,优化后的菊苣叶黄酮的提取条件为液料比37.0 mL/g、提取时间59.2 min、提取温度43℃。此条件下菊苣叶黄酮得率为3.09%,与预测值基本一致,表明此优化方法合理可靠。体外抗氧化活性试验显示,菊苣叶黄酮对1,1-二苯基-2-三硝基苯肼(DPPH)自由基、2,2-联氮-二(3-乙基-苯并噻唑-6-磺酸)二铵盐(ABTS+)自由基的清除能力与浓度呈计量关系。研究表明,菊苣叶黄酮的抗氧化能力较强,具有作为饲料添加剂开发的潜力。 The study was to enhance the feed value of chicory leaves.Through response surface methodology,the experiment optimized the extraction process of flavonoids from chicory leaves and evaluates their antioxidant activity.Based on single-factor experiments,the design and optimization of the Box-Behnken design were conducted with the response variable being the yield of flavonoids,to analyze the effects of the liquid-to-solid ratio,extraction temperature,and extraction time on the yield of flavonoids in chicory leaves.The results showed that the optimized conditions for the extraction of chicory leaf flavonoids were a liquid-to-solid ratio of 37.0 mL/g,an extraction time of 59.2 minutes,and an extraction temperature of 43℃.Under these conditions,the yield of chicory leaf flavonoids was 3.09%,which was basically consistent with the predicted value,indicating that this optimization method was reasonable and reliable.In vitro antioxidant activity tests showed that the scavenging ability of chicory leaf flavonoids on DPPH and ABTS+radicals was concentration-dependent.The study suggests that chicory leaf flavonoids have strong in vitro antioxidant capacity and potential for feed additive development.
