高效液相色谱法测定化妆品中10-羟基-2-癸烯酸

Determination of 10-hydroxy-2-decenoic acid in cosmetics by high performance liquid chromatography

建立高效液相色谱测定含有蜂王浆成分化妆品中10-羟基-2-癸烯酸(10-HDA)的方法。在1.000 g样品中加入8 mL甲醇,超声提取10 min,静置至室温,用甲醇定容至10 mL,以10000 r/min转速离心5 min,上清液经0.45μm滤膜过滤,在35℃柱温下,采用C_(18)柱(250 mm×4.6 mm,5μm)进行分离。以甲醇-0.12%磷酸溶液(体积比为55∶45)为流动相进行等度洗脱,流量为1.0 mL/min,进样体积为10μL,以二极管阵列检测器检测,检测波长为210 nm。10-HDA的质量浓度在0.22~54.19μg/mL范围内与色谱峰面积线性关系良好,相关系数为0.9999,方法检出限和定量限分别为0.65、2.17μg/g。样品加标回收率为96.59%~102.09%,测定结果的相对标准偏差为0.43%~1.33%(n=6)。该方法高效,灵敏度高,可用于含蜂王浆成分化妆品中10-HDA的含量检测。

A method for the determination of 10-hydroxy-2-decenoic acid(10-HDA)in cosmetics containing royal jelly by high performance liquid chromatography was established.The 1.000 g sample was added with 8 mL methanol,ultrasonically extracted for 10 min,then methanol volume was fixed to 10 mL at room temperature,and centrifuged at 10000 r/min for 5 min.The supernatant was filtered by 0.45μm filter membrane and separated by C_(18) column(250 mm×4.6 mm,5μm)at the column temperature of 35℃.Methanol-0.12%phosphoric acid solution(volume ratio was 55∶45)was used as the mobile phase for isocratic elution.The flow rate and the sample volume were respectively 1.0 mL/min and 10μL,and the detection wavelength was 210 nm with diode array detector.The mass concentration of 10-HDA had a good linear relationship with the chromatographic peak area in the range of 0.22-54.19μg/mL,and the correlation coefficient was 0.9999.The limit of detection and limit of quantification were 0.65μg/g and 2.17μg/g,respectively.The recoveries of sample spiked were 96.59%-102.09%,and the relative standard deviations of the determination results were 0.43%-1.33%(n=6).The method is efficient and sensitive,which can be used for the determination of 10-HDA in cosmetics containing royal jelly.
