HPLC-MS/MS测定化妆品中羟丙基四氢吡喃三醇的含量及构型比例

Determination of Hydroxypropyl Tetrahydropyrantriol in Cosmetics by HPLC-MS/MS and Configuration Ratio

摘要 建立了一种高效液相色谱-串联质谱法,能够同时测定化妆品中羟丙基四氢吡喃三醇(玻色因)含量及其不同构型的比例。试样经四氢呋喃溶解混匀后,超声提取,使用氨基柱洗脱分离,选择电喷雾离子源,以MRM模式采集进行分析,外标法定量,利用峰面积计算(β, S)构型玻色因与(β, R)构型玻色因的比例。结果表明,羟丙基四氢吡喃三醇,在10~200 mg/L范围内,浓度与响应值呈显著线性关系,相关系数(r)≥0.999,回收率范围均在86.6%~93.1%之间,相对标准偏差(n=6)小于2.6%,方法定量限为1 000 mg/kg。该方法操作简便,针对高极性、结构差异微小的化合物具有较好的保留和分离效果,且灵敏度高、专属性强、准确性好,前处理操作简易,能够进行化妆品中羟丙基四氢吡喃三醇含量及其不同构型比例的大批量测定。 A high performance liquid chromatography-tandem mass spectrometry(HPLC-MS/MS)method was established to simultaneously determine the content of hydroxypropyl tetrahydropyrantriol(Proxylane)and the proportion of different configurations in cosmetics.The sample was dissolved and mixed with tetrahydrofuran,and then extracted by ultrasonic extraction.The amino column was used for positive phase elution separation.The electrospray ion source was selected,and the MRM mode was collected for analysis.The external standard method was used for quantification,and the peak area was used to calculate the ratio of(β,S)configuration Proxylane and(β,R)configuration Proxylane.The results showed that there was a significant linear relationship between the concentration and the response value of hydroxypropyl tetrahydropyrantriol in the range of 10~200 mg/L,the correlation coefficient(r)≥0.999,the recovery range was between 86.6%~93.1%,the relative standard deviation(n=6)was less than 2.6%,and the quantitative limit of the method was 1000 mg/kg.The method was simple to operate and had good retention and separation effects for compounds with high polarity and small structural differences.It had high sensitivity,strong specificity,good accuracy and simple pretreatment operation.It can be used for mass determination of hydroxypropyl tetrahydropyrantriol content and its different configuration ratios in cosmetics.
