烟草青枯病根际土壤细菌群落动态变化分析

Analysis of dynamic changes of rhizosphere bacterial communities in the pathogenesis of tobacco bacterial wilt

摘要 为了解山东烟草种植区烟草青枯病病害发生过程中的土壤微生物组群落动态变化,通过高通量测序,系统分析了山东沂水县地区烟草根际微生物组。扩增子测序分析表明,与移栽前相比,青枯病发病初期根际土壤微生物多样性显著降低,但是随着病害的发展,后续根际土壤微生物多样性又有恢复趋势;进一步对比移栽前和青枯病病发初期烟草根际微生物组成的差异,探究了整个病害发生的动态过程与根际细菌群落变化之间的规律,并通过构建相关性网络发现了小粒胞菌属(Granulicella)、无色杆菌属(Achromobacter)、出芽单胞菌属(Gemmatimonas)、居土杆菌属(Humibacter)、弯曲杆状菌属(Flexivirga)、束缚菌属(Conexibacter)与青枯病的发生存在潜在的作用关系。综上所述,本研究通过时间维度分析烟草发病动态过程根际细菌组成的差异,发现了潜在的烟草青枯病防治菌属以及预警菌属,为后续山东烟草土传病害预警和防控奠定了基础。 In order to understand the dynamic changes of soil microbial community during the whole disease process of tobacco bacterial wilt in cultivation areas of tobacco of Shandong Province,this study systematically analyzed the tobacco rhizosphere microbial community in Yishui County,Shandong Province by high-throughput sequencing.Amplicon sequencing analysis showed that,compared with before transplanting,the microbial diversity of rhizosphere soil significantly decreased when tobacco bacterial wilt was initially manifested,but there was a trend of recovery in the subsequent rhizosphere soil microbial diversity with the development of the disease.The microbial composition in tobacco rhizosphere before transplanting and early onset of tobacco bacterial wilt were compared,and the rule between the dynamic process of the whole disease occurrence and the change of rhizosphere bacterial community was explored.Via a constructed correlation network,Granulicella,Achromobacter,Gemmatimonas,Humibacter,Flexivirga and Conexibacter were detected to have potential effect on the occurrence of bacterial wilt.In summary,the dfference of rhizosphere bacteria composition in a time-dimensional process of tobacco onset was analyzed,and the potential bacteria genera with disease control and warning potentials were found,which laid a foundation for the early warning and control of soil-borne tobacco diseases in Shandong.
