基于酪氨酸酶抑制活性和DPPH自由基清除活性的逍遥散组方研究

Study on Xiaoyao Powder formula based on tyrosinase inhibitory activity and DPPH free radical scavenging activity

摘要 以逍遥散为主要研究对象,以1,1-二苯基-2-三硝基苯肼(DPPH)自由基清除活性和酪氨酸酶抑制活性的综合评价为指标,对其水煎液的美白功效进行了初步评价,并运用均匀设计法对逍遥散的组方进行了研究。结果表明,逍遥散水煎液对DPPH自由基具有一定的清除能力,当其水煎液浓度为0.1g/mL时,其清除率为91.09%;对酪氨酸酶的抑制活性则与其水煎液的质量浓度有关,表现为高浓度时抑制,低浓度时激活,当其水煎液浓度为0.1g/mL时,其对酪氨酸酶的抑制率为48.73%;在逍遥散美白功效评价中,方中煨姜和薄荷对该方影响较小,在新组方研究中以柴胡、当归、白芍、白术、茯苓和甘草六味药为主要组成;在均匀设计试验中,当逍遥散中m(柴胡)∶m(当归)∶m(白芍)∶m(白术)∶m(茯苓)∶m(甘草)的比例约为1.5∶1.7∶2.2∶2.1∶2.1∶1时,具有较好的美白效果。 Xiaoyao Powder was taken as the main study object,the whitening efficacy of its decoction was evaluated by the comprehensive evaluation of DPPH(1,1-diphenyl-2-picryl-hydrazy)free radical scavenging activity and tyrosinase inhibiting activity.Then the composition of Xiaoyao Powder was studied by uniform design method.Results showed that,the extractive of Xiaoyao Powder displayed DPPH free radical scavenging capacity,and When the concentration of its aqueous solution was 0.1g/mL,the clearance rate was 91.09%.With respect to activity of tyrosinase,it related to mass concentration,and it displayed inhibitory action at high mass concentration,while at lower mass concentration it displayed activation effect,and When the concentration of its aqueous solution was 0.1g/mL,the inhibition rate of tyrosinase was 48.73%.There was little effect with the presence or absence of simmered ginger and peppermint in the evaluation of whitening efficacy,so the new formula was mainly composed of Chaihu(Radix bupleuri),Danggui(Angelicae Sinensis Radix),Baishao(Radix Paeoniae Pall),Baizhu(Atractylodis Macrocephalae Rhizoma),Fuling(Poria cocos)and Gancao(Glycyrrhizae Radix et Rhizoma).In the uniform designed experiment,the results showed that when the mass ratio in the composition of the new Xiaoyao Powder Chaihu(Radix bupleuri),Danggui(Angelicae Sinensis Radix),Baishao(Radix Paeoniae Pall),Baizhu(Atractylodis Macrocephalae Rhizoma),Fuling(Poria cocos)and Gancao(Glycyrrhizae Radix et Rhizoma)was about 1.5∶1.7∶2.2∶2.1∶2.1∶1,the new Xiaoyao Powder had a better whitening effect.
