油菜花粉玛咖咀嚼片的制备及槲皮素和山奈酚的含量测定

摘要 目的:探究油菜花粉玛咖咀嚼片的制备工艺及槲皮素和山奈酚含量测定的方法。方法:通过湿法制粒工艺,制备油菜花粉玛咖咀嚼片;采用高效液相色谱(HPLC)法对油菜花粉玛咖咀嚼片中槲皮素和山奈酚的含量进行测定,色谱柱为ODS-C_(18)柱,流动相为甲醇-水,进行等度洗脱,体积流量为1.0 mL/min;柱温为35℃,检测波长为370 nm,进样量为5μL。结果:油菜花粉玛咖咀嚼片的制备工艺为油菜花粉∶玛咖提取物粉末∶填充剂=1∶1∶2,填充剂(微晶纤维素:木糖醇:乳糖=6∶4∶3)。所得咀嚼片表面光洁、甜度适中且质量稳定可控。槲皮素和山奈酚的线性范围分别为2.29~45.83μg/mL、3.75~75μg/mL,相关系数(r)分别为0.9996和0.9992;平均加样回收率分别为99.79%和101.02%,RSD分别为3.33%和2.56%。结论:油菜花粉玛咖咀嚼片制备工艺稳定可行,槲皮素和山奈酚含量测定方法准确可靠,可用于该咀嚼片的质量控制。 Objective To establish the preparation process of rape pollen maca chewable tablets and the determination of quercetin and kaempferol contents.Methods Rape pollen maca chewable tablets were prepared by wet granulation process.Determination of quercetin and kaempferol in Rapeseed Pollen Maca Chewable Tablets by High Performance Liquid Chromatography(HPLC),the chromatographic column is an ODS-C_(18)column,and the mobile phase is methanol water for isocratic elution,with a volume flow rate of 1.0 mL/min,the column temperature is 35℃,the detection wavelength is 370 nm,and the injection amount is 5μL.Results The preparation procedure of rapeseed pollen Maca chewable tablets was as follows:Rape pollen:Maca extract powder:filler ratio is 1∶1∶2,The ratio of microcrystalline cellulose to xylitol to lactose in the filler is 6∶4∶3.Wet granulation and tablet pressing were carried out.The obtained chewable tablets have a smooth surface,moderate sweetness,and stable and controllable quality.A method for the determination of quercetin and kaempferol was established.The linear ranges of quercetin and kaempferol were 2.29-45.83μg/mL and 3.75-75μg/mL,respectively,and the correlation coefficients were 0.9996 and 0.9992,respectively.The average recoveries of quercetin and kaempferol were 99.79%and 101.02%,respectively.Conclusion The preparation procedure of rapeseed pollen Maca chewable tablets is stable and feasible.The method for the determination of quercetin and kaempferol is accurate and reliable and can be used for the quality control of the chewable tablets.
