摘要
目的研究艾灸对豚鼠血清肿瘤坏死因子-α(TNF-α)、白细胞介素6(IL-6)、C-C模体趋化因子配体22(CCL22)表达的影响。方法25只豚鼠取5只作为空白对照组,其余作为治疗组,按照随机数字表法将剩余20只豚鼠根据不同时间点分为4组:艾灸治疗后即刻组(T-1组)、艾灸治疗后1个月组(T-2组)、艾灸治疗后2个月组(T-3组)、艾灸治疗后3个月组(T-4组),每组各5只。各组豚鼠背部脱毛约2 cm×2 cm,除空白对照组外,其余4组豚鼠均进行艾灸,艾灸治疗穴为经外奇穴“灸癜风”和“阿是穴”,“阿是穴”即脱毛区。穴上3 cm处悬灸,采用温和灸补法,每穴约10 min/次,1次/d,疗程为1个月。空白对照组仅采用同法固定,不进行艾灸。比较5组豚鼠血清TNF-α、IL-6、CCL22表达水平。结果T-1组、T-2组、T-4组豚鼠血清TNF-α表达水平分别为(42.49±4.66)、(88.75±23.18)、(79.76±20.20)ng/L,均较空白对照组[(119.55±11.25)ng/L]明显降低,差异均有统计学意义(P<0.05);T-3组豚鼠血清TNF-α表达水平与空白对照组比较,差异无统计学意义(P>0.05)。T-1组豚鼠血清IL-6表达水平为(37.70±8.94)ng/L,较空白对照组[(51.51±9.95)ng/L]显著降低,差异有统计学意义(P<0.05);T-2组、T-3组、T-4组豚鼠血清IL-6表达水平与空白对照组比较,差异无统计学意义(P>0.05)。T-1组、T-2组、T-3组、T-4组豚鼠血清CCL22表达水平分别为(176.14±27.62)、(197.06±34.05)、(146.22±9.85)、(216.52±38.65)ng/L,均较空白对照组显著升高[(120.86±23.89)ng/L],差异均有统计学意义(P<0.05)。结论艾灸可降低豚鼠血清TNF-α、IL-6水平,升高豚鼠血清CCL22水平,且对TNF-α、CCL22的影响可持续到艾灸结束后3个月。
Objective To study the effect of moxibustion on the expression of serum tumor necrosis factor-α(TNF-α),interlukin-6(IL-6),C-C motif chemokine ligand 22(CCL22)in guinea pigs.Methods Five of the 25 guinea pigs were taken as the blank control group,and the rest were taken as the treatment group.According to the random number table method,the remaining 20 guinea pigs were divided into 4 groups according to different time points,which were immediately after moxibustion treatment group(T-1 group),1 month after moxibustion treatment group(T-2 group),2 months after moxibustion treatment group(T-3 group),and 3 months after moxibustion treatment group(T-4 group),with 5 guinea pigs in each group.The back depilation of guinea pigs in each group was about 2 cm×2 cm.Except for the blank control group,the other four groups of guinea pigs were treated with moxibustion.The moxibustion treatment points were"Jiudianfeng"and"Ashi"."Ashi acupoint"is the depilation area.Hanging moxibustion at 3cm above acupoints adopts mild moxibustion and tonifying method,each acupoint is about 10min once a day,and the course of treatment is 1 month.The blank control group was only fixed by the same method without moxibustion.The expression levels of serum TNF-α,IL-6 and CCL22 in the five groups were compared.Results The expression levels of serum TNF-αin T-1 group,T-2 group and T-4 group were(42.49±4.66),(88.75±23.18)and(79.76±20.20)ng/L,respectively,which were significantly lower than those in blank control group[(119.55±11.25)ng/L],and the differences were statistically significant(P<0.05).There was no significant difference in the expression level of serum TNF-αbetween the T-3 group and the blank control group(P>0.05).The expression level of serum IL-6 in the T-1 group was(37.70±8.94)ng/L,which was significantly lower than that in the blank control group[(51.51±9.95)ng/L],and the difference was statistically significant(P<0.05).There was no significant difference in serum IL-6 expression level between T-2 group,T-3 group,T-4 group and blank control group(P>0.05).The expression levels of CCL22 in serum of guinea pigs in T-1 group,T-2 group,T-3 group and T-4 group were(176.14±27.62),(197.06±34.05),(146.22±9.85)and(216.52±38.65)ng/L,respectively,which were significantly higher than those in the blank control group[(120.86±23.89)ng/L],and the differences were statistically significant(P<0.05).Conclusion Moxibustion can decrease the level of serum TNF-α、IL-6 in guinea pigs whereas increasethe level of serum CCL22 in guinea pigs,and the effect of moxibustion on TNF-αCCL22 can last 3 months after the end of moxibustion.
