摘要
为了解枸杞果实中的活性成分在不同光照条件下合成和积累的变化,研究黄酮类物质和类胡萝卜素合成的影响及调控机制,将枸杞果实置于紫外线(UV)辐射下,并进行转录组测序分析。研究发现,适度的紫外辐射显著增加了枸杞果实中黄酮类和类胡萝卜素的含量,其中黄酮合成相关基因CHS、HCT、F3H和FLS,以及类胡萝卜素合成相关基因PSY和CYP707A的表达均显著上调。GO和KEGG通路分析表明,这些基因主要参与细胞代谢和信号传导过程,并促进了生松素查尔酮、二氢山柰酚、斛皮黄酮、杨梅酮及番茄红素的合成。本研究为利用紫外辐射调控枸杞活性成分的合成提供了科学依据。
In order to understand the changes in the synthesis and accumulation of active components in Lycium barbarum fruit under different light conditions,and to study the effects on flavonoids and carotenoids synthesis and the regulatory mechanisms,Lycium barbarum fruit was subjected to ultraviolet(UV)radiation and analyzed by transcriptome sequencing.It was found that moderate UV radiation significantly increased the content of flavonoids and carotenoids in Lycium barbarum fruits,in which the expression of flavonoid synthesis-related genes CHS,HCT,F3H and FLS,as well as the carotenoid synthesis-related genes PSY and CYP707A,was significantly up-regulated.The analysis of the GO and KEGG pathways indicated that these genes were mainly involved in cellular metabolism and signaling processes,and promoted the synthesis of raw pinein chalcone,dihydrokaempferol,dendrobium flavonoids,populinone and lycopene.This study provides a scientific basis for the use of UV radiation to regulate the synthesis of active ingredients in Lycium barbarum.
