摘要
目的探讨河蚌多糖水提取物体外抗肿瘤活性。方法通过离心、浓缩和冷冻干燥的方法制备出可溶性的河蚌多糖水提物,分别采用CCK-8实验、细胞划痕实验和Transwell迁移实验检测河蚌多糖水提取物对肝癌细胞Hep3B、恶性黑色素瘤细胞A375和肺癌细胞PC9增殖、迁移能力的抑制作用。结果CCK-8实验表明,河蚌多糖水提取物具有时间-浓度依赖性抑制上述3种恶性肿瘤细胞的增殖;划痕实验和Transwell迁移实验结果表明,与对照组相比,河蚌多糖水提取物能够有效抑制上述3种恶性肿瘤细胞的迁移(P均<0.05)。结论河蚌多糖水提取物可以有效抑制多种恶性肿瘤细胞增殖和迁移,为其后续开发和应用提供一定的参考价值。
Objective To investigate the antitumor activity of mussel polysaccharide water extract in vitro.Methods Mussel polysaccharide water extract was prepared by centrifugating,concentrating and freeze-drying.Then CCK-8 assay,cell scratch assay and Transwell migration assay were used to detect the inhibitory effects of the mussel polysaccharide water extract on the proliferation and migration of hepatoma carcinoma cell Hep3B,malignant melanoma cell A375 and lung carcinoma cell PC9 respectively.Results The CCK-8 assay result showed that the mussel polysaccharide water extract inhibited the proliferation of the above three kinds of malignant tumor cells in a time and concentration dependent manner.The scratch assay and Transwell migration assay results showed that the mussel polysaccharide water extract effectively inhibited the migration of the above three kinds of malignant tumor cells compared with the control group(all P<0.05).Conclusion The mussel polysaccharide water extract can effectively inhibit the proliferation and migration of malignant tumor cells,providing a certain reference value for its subsequent development and application.
