聚赖氨酸处理下养殖大黄鱼冷藏品质及微生物多样性变化

摘要 为探究聚赖氨酸(ε-Polylysine,PL)对养殖大黄鱼的保鲜效果,通过测定色泽、质构、pH、硫代巴比妥酸反应物(TBARS)值、总挥发性盐基氮(TVB-N)含量、菌落总数等指标,并结合Illumina MiSeq高通量测序技术,研究不同质量浓度(0.05、0.10、0.15、0.20 g/L)PL处理对养殖大黄鱼在冷藏(4℃)期间品质及微生物多样性的影响。结果表明:PL处理可有效延缓大黄鱼在冷藏过程中L^(*)值、硬度、弹性的下降以及a^(*)值、b^(*)值、pH、TBARS值、TVB-N含量、菌落总数的上升,色泽、质构劣变减缓,脂肪氧化、蛋白质分解及微生物繁殖得到有效抑制,且PL处理浓度越高其保鲜效果越明显,0.20 g/L处理组冷藏期较对照组(蒸馏水处理)延长2 d。测序结果显示,0.20 g/L PL处理下,鱼肉冷藏期间菌群多样性降低,假单胞菌属、希瓦氏菌属、不动杆菌属等腐败菌属相对丰度下降。说明PL处理可有效抑制腐败菌生长,进而延缓冷藏期间鱼体品质劣变进程,延长货架期,是潜在的大黄鱼冷藏保鲜方法。 To explore the effect of ε-polylysine(PL)on the quality and microbial diversity of farmed large yellow croaker(Larimichthys crocea)under refrigeration(4℃),the fresh samples were subjected to PL solutions at different concentrations(0.05,0.10,0.15,0.20 g/L).The changes in color,texture,pH,thiobarbituric acid reactive substances(TBARS),total volatile base nitrogen(TVB-N)content,and total viable count during refrigerated storage were evaluated.In addition,the microbial diversity was analyzed by Illumina MiSeq high-throughput sequencing technology to confirm the preservative mechanism of PL on the refrigerated fish.The results showed that PL treatment could effectively delay the decrease of L^(*)value,hardness,elasticity,as well as the increase of a^(*)value,b^(*)value,pH,TBARS,TVB-N content,and total bacterial count of large yellow croaker during refrigeration,exhibiting an effective inhibition on the color changes,texture deterioration,lipid oxidation,protein decomposition,and microbial reproduction.With the increase of PL concentration,the effect became more obvious.Compared with the control gracp(treated with distilled water),the shelf life of samples could be prolonged by 2 days under the treatment of 0.20 g/L PL.The sequencing results revealed that PL(0.20 g/L)could decrease microbial diversity and richness,especially,Pseudomonas,Shewanella,and Acinetobacter in samples.PL can effectively inhibit the growth of spoilage bacteria in refrigerated farmed large yellow croaker,thereby delaying its quality deterioration process and extending its shelf life.This study indicates that PL treatment is a potential method for the preservation of refrigerated large yellow croaker.
