摘要
目的:观察益母缩宫颗粒对不全流产大鼠子宫中辅助T细胞1/辅助T细胞2(Th1/Th2)亚群及相关收缩因子的影响,以探讨其改善药物流产后异常阴道流血的作用机制。方法:以大鼠为研究对象,随机选取9只雌鼠为空白组,剩余雌鼠与成熟雄鼠合笼制备早孕不全流产模型,将造模成功的雌鼠分为模型组10只,对照组9只,益母缩宫颗粒低剂量组10只、中剂量组9只、高剂量组9只。空白组、模型组灌胃动物饮用水每天1 mL/100 g,连续7 d;益母缩宫颗粒低、中、高剂量组分别每天灌胃益母缩宫颗粒1.56 g/kg、3.13 g/kg、6.25 g/kg,连续7 d。对照组每天注射缩宫素0.9 U/kg,连续3 d。观察大鼠干扰素-γ(IFN-γ)、白细胞介素4(IL-4)、前列腺素F2α(PGF2α)水平及间隙连接蛋白43(CX43)的表达。结果:模型组IFN-γ、IL-4、PGF2α与空白组比较,益母缩宫颗粒低剂量组IFN-γ、IL-4、PGF2α及中、高剂量组IL-4与对照组比较,低剂量组IFN-γ、PGF2α及中、高剂量组和对照组的IFN-γ、IL-4、PGF2α与模型组比较,差异均有统计学意义(P<0.01)。模型组IFN-γ/IL-4与空白组比较,益母缩宫颗粒低、中、高剂量组及对照组IFN-γ/IL-4与模型组比较,低、中、高剂量组IFN-γ/IL-4与对照组比较,差异均有统计学意义(P<0.01)。模型组及各用药组CX43 OD值与空白组比较,中、高剂量组及对照组CX43 OD值与模型组比较,低剂量组CX43 OD值与对照组比较,差异均有统计学意义(P<0.01)。结论:益母缩宫颗粒可能通过调控Th1/Th2亚群向Th1偏移而引起母-胎之间的免疫排斥,促进残留蜕膜组织、绒毛组织变性、坏死,减少不全流产后宫内残留,其作用机制可能与促进子宫平滑肌收缩有关。
Objective:To investigate the effect of Yimu Suogong Granule on T-helper type 1/T-helper type 2(Th1/Th2)subsets and related contraction factors in the uterus of rats with incomplete medical abortion,as well as the mechanism of action of Yimu Suogong Granule in improving abnormal vaginal bleeding after medical abortion.Methods:A total of 9 female rats were selected as blank group,and the remaining female rats were caged with mature male rats to establish a model of incomplete abortion during early pregnancy.After successful modeling,the female rats were divided into model group with 10 rats,control group with 9 rats,low-dose Yimu Suogong Granule group with 10 rats,middle-dose Yimu Suogong Granule group with 9 rats,and high-dose Yimu Suogong Granule group with 9 rats.The rats in the blank group and the model group were given animal drinking water 1 mL/100 g by gavage for 7 consecutive days,and those in the low-,middle-,and high-dose Yimu Suogong Granule groups were given Yimu Suogong Granule by gavage at a dose of 1.56 g/kg,3.13 g/kg,and 6.25 g/kg,respectively,by gavage for 7 consecutive days.The rats in the control group were given injection of oxytocin 0.9 U/kg for 3 consecutive days.The rats were observed in terms of the expression of interferon-γ(IFN-γ),interleukin-4(IL-4),prostaglandin F2α(PGF2α),and connexin 43(CX43).Results:There were significant differences in IFN-γ,IL-4,and PGF2αbetween the model group and the blank group and between the low-dose Yimu Suogong Granule group and the control group,and there was a significant difference in IL-4 between the middle-and high-dose Yimu Suogong Granule groups and the control group;there were significant differences in IFN-γand PGF2αbetween the low-dose Yimu Suogong Granule group and the model group,and there were significant differences in IFN-γ,IL-4,and PGF2αbetween the middle-and high-dose Yimu Suogong Granule groups and the model group and between the control group and the model group(P<0.01).There was a significant difference in IFN-γ/IL-4 between the model group and the blank group,between the low-,middle-,and high-dose Yimu Suogong Granule groups and the model group,between the control group and the model group,and between the low-,middle-,and high-dose Yimu Suogong Granule groups and the control group(P<0.01).There was a significant difference in CX43 OD value between the model group and each administration group,between the middle-and high-dose Yimu Suogong Granule groups and the model group,between the control group and the model group,and between the low-dose Yimu Suogong Granule group and the control group(P<0.01).Conclusion:Yimu Suogong Granule can cause maternal-fetal immunological rejection by regulating the shift of Th1/Th2 subsets to Th1,promote the degeneration and necrosis of residual decidual tissue and chorion tissue,and reduce intrauterine residues after incomplete abortion,possibly by promoting the contraction of uterine smooth muscle.
