低温胁迫下西瓜嫁接苗的蛋白质组学分析

摘要 为了研究西瓜嫁接苗遭遇到低温胁迫后,蛋白表达丰度的变化,深入了解西瓜苗期耐冷响应过程中所涉及的代谢途径,探讨相关的分子调控机理。本研究对西瓜嫁接苗和自根苗进行低温处理,采用iTRAQ技术对其进行蛋白质组学的鉴定和定量分析,然后利用数据库进行比对,并采用GO及KEGG注释其功能和代谢通路,最终获得了差异蛋白985个,其中上调蛋白545个,下调蛋白440个,涉及以下4种主要类别的蛋白质:碳水化合物代谢(26.52%)、遗传信息翻译(19.78%)、能量代谢(17.83%)和氨基酸代谢(13.70%)。蛋白质组学分析显示,与自根苗相比,嫁接苗低温耐受性的增加与甘油醛-3-磷酸脱氢酶(GAPDH)和核酮糖二磷酸羧化酶(Rubisco)的积累有关。 To investigate the changes in protein expression abundance of grafted watermelon seedlings under low temperature stress,and gain deeper understanding of the metabolic pathways involved in the cold tolerance response process during watermelon seedling stage,as well as to explore the relevant molecular regulatory mechanisms,low-temperature treatment was conducted on both grafted and self rooted seedlings for identification and quantitative analysis using iTRAQ technology.Then,the obtained peptides and proteins were analyzed using databases,and their functions and metabolic pathways were annotated using GO and KEGG.In total,985 differential proteins were identified,including 545 upregulated proteins and 440 downregulated proteins.These proteins were categorized into four main groups:carbohydrate metabolism(26.52%),genetic information translation(19.78%),energy metabolism(17.83%),and amino acid metabolism(13.70%).Proteomics analysis showed that the enhanced low temperature tolerance observed in grafted seedlings was related to the accumulation of Glyceraldehyde 3-phosphate dehydrogenase(GAPDH)and Ribulose bisphosphate carboxylase(Rubisco)compared to self-rooted seedlings.The results of this study provide a scientific basis for grafting to enhance the cold tolerance of watermelon seedlings.
