幼儿配方羊奶粉对抗生素所致腹泻小鼠的影响

摘要 文章探讨幼儿配方羊奶粉对抗生素所致腹泻小鼠的影响,为幼儿配方羊奶粉的推广应用提供依据。将50只KM小鼠按性别体质量随机分为空白对照组、模型对照组、羊奶粉低剂量组、羊奶粉高剂量组、幼儿配方牛奶粉组,每组10只,雌雄各半。奶粉组按20 mL/kg体质量灌胃,空白和模型对照组灌胃纯水,每天2次,连续14 d。给样7 d后,空白对照组灌胃生理盐水,其余各组灌胃头孢拉定(1.00 g/kg体质量)+庆大霉素(0.16 g/kg体质量),0.4 mL/只,2次/d,连续4 d,给抗生素2 h后继续给受试样品,试验结束前1天收集小鼠粪便,用于测定粪便含水量及肠道微生物分析,末次给样后次日眼眶采血测定血清TNF-α、IL-2、IL-6、IL-1β水平,取空肠、结肠组织进行组织病理学检查。结果表明,与空白对照组比较,模型对照组动物粪便含水量显著升高(P<0.05),血清IL-2水平极显著降低(P<0.01),IL-6、IL-1β水平极显著升高(P<0.01),空肠及回肠组织内可见隐窝数量减少,肠绒毛变短,固有层炎性细胞浸润等病理改变,肠道菌群结构改变,多样性降低;与模型对照组比较,羊奶粉高剂量组及幼儿配方牛奶粉组动物粪便含水量显著或极显著降低(P<0.05或P<0.01),血清IL-2水平显著升高(P<0.05),IL-6水平显著下降(P<0.05);羊奶粉对小鼠空肠和结肠组织病变有明显修复作用,对抗菌素所致小鼠肠道菌群多样性及组成有显著调节功能。幼儿配方羊奶粉可能通过提高肠道免疫功能,调节肠道菌群组成改善抗生素致腹泻小鼠肠道病理损伤。 To investigate the effect of infant sheep formula milk powder on antibiotic-induced diarrhea in mice,and to provide experimental basis for the promotion and application of infant sheep formula milk powder.50 KM mice were randomly divided into blank control group,model control group,sheep milk powder low dose group,high dose group and infant formula milk powder group according to gender weight,10 mice in each group,half male and half female.The milk powder group was given 20 mL/kg BW gavage,and the blank and model control groups were given gavage with pure water twice a day for 14 consecutive days.After 7 days of sample administration,the blank control group was given gavage of normal saline,and the other groups were given gavage of cefradine(1.00 g/kg BW)+gentamicin(0.16 g/kg BW),0.4 mL/only,twice a day for 4 consecutive days.After 2 h of antibiotics,the samples were continued to be given.It was used for the determination of fecal water content and intestinal microbial analysis.The next day after the last sample was given,the levels of serum TNF-α,IL-2,IL-6 and IL-1βwere measured by orbital blood collection.The jejunum and colon tissues were taken for histopathological examination.Result showed that compared with blank control group,fecal water content in model control group was significantly increased(P<0.05),serum IL-2 level was significantly decreased(P<0.01),IL-6 and IL-1βlevels were significantly increased(P<0.01),the number of crypts in jejunum and ileum tissue was decreased,intestinal villi was shorter,lamina propria inflammatory cell infiltration and other pathological changes were observed.The structure of intestinal flora changed and the diversity decreased.Compared with model control group,fecal water content in sheep milk powder high-dose group and infant formula milk powder group was significantly decreased(P<0.05 or P<0.01),serum IL-2 level was significantly increased(P<0.05),and IL-6 level was significantly decreased(P<0.05).Sheep milk powder can obviously repair jejunum and colonic tissue lesions in mice,and regulate the diversity and composition of intestinal flora caused by antibiotics.Infant sheep formula milk powder may improve intestinal pathological injury of antibiotic-induced diarrhea mice by improving intestinal immune function and regulating intestinal flora composition.
