摘要
目的分析H.pylori阳性胃癌患者临床病理参数与miR-191/磷脂酶C-delta-1(phospholipase C-delta-1,PLCd1)表达的关系。方法选取2021年1月至2022年12月收治的H.pylori阳性胃癌患者86例作为H.pylori组,80例H.pylori阴性胃癌患者作为无H.pylori组。回顾性分析两组患者临床资料。比较胃癌组织与癌旁组织miR-191、PLCd1表达及两者比值miR-191/PLCd1在两组间的差异;分析胃癌组织miR-191、PLCd1表达在H.pylori阳性胃癌患者中的相关性;比较并分析H.pylori阳性胃癌患者不同临床病理特征与癌组织miR-191/PLCd1比值的差异和相关性。结果H.pylori组和无H.pylori组癌组织miR-191相对表达量及miR-191/PLCd1比值均显著高于其癌旁组织(P<0.05),H.pylori组癌组织miR-191相对表达量及miR-191/PLCd1比值高于无H.pylori组癌组织(P<0.05);H.pylori组和无H.pylori组癌组织PLCd1相对表达量均明显低于其癌旁组织(P<0.05),H.pylori组癌组织PLCd1相对表达量低于无H.pylori组癌组织(P<0.05)。Pearson相关系数分析显示,癌组织miR-191相对表达量与PLCd1相对表达量呈负相关(r=-0.649,P<0.05)。H.pylori阳性胃癌患者中,浸润深度T_(3)~T_(4)者癌组织miR-191/PLCd1比值明显高于T_(1)~T_(2)者(P<0.05),淋巴结转移者癌组织miR-191/PLCd1比值明显高于无淋巴结转移者(P<0.05),Spearman秩相关性分析显示,癌组织miR-191/PLCd1比值与浸润深度、淋巴结转移呈正相关(r=0.775、0.728,P<0.05)。结论癌组织miR-191/PLCd1比值与H.pylori阳性胃癌生物学行为密切相关,有望成为胃癌诊疗的新靶点。
Objective To analyze the relationship between clinicopathological parameters and expressions of miR-191/phospholipase C delta 1(PLCd1)in patients with H.pylori-positive gastric cancer.[WTHZ]Methods A total of 86 patients with H.pylori-positive gastric cancer from Jan.2021 to Dec.2022 were selected as H.pylori group,and 80 patients with H.pylori-negative gastric cancer were enrolled as non-H.pylori group.Clinical data of patients in both groups were retrospectively analyzed,and the differences in expressions of miR-191,PLCd1 and miR-191/PLCd1 ratio were compared between gastric cancer tissues and adjacent tissues.The correlation between expression of miR-191 and expression of PLCd1 in gastric cancer tissues in patients with H.pylori-positive gastric cancer was analyzed.The difference and the correlation between miR-191/PLCd1 ratio and clinicopathological parameters in patients with H.pylori-positive gastric cancer were analyzed.[WTHZ]Results The relative expression level of miR-191 and miR-191/PLCd1 ratio in cancer tissues in H.pylori group and non-H.pylori group were significantly higher than those in adjacent tissues(P<0.05),furthermore,the relative expression level of miR-191 and the miR-191/PLCd1 ratio in cancer tissues of H.pylori group were higher than those in cancer tissues in non-H.pylori group(P<0.05).The relative expression level of PLCd1 in cancer tissues in H.pylori group and non-H.pylori group was significantly lower than that in adjacent tissues(P<0.05),and the relative expression level of PLCd1 in cancer tissues of H.pylori group was lower than that in non-H.pylori group(P<0.05).Pearson correlation coefficient analysis showed that the relative expression level of miR-191 in cancer tissues was significantly negatively correlated with relative expression level of PLCd1(r=-0.649,P<0.05).Among patients with H.pylori-positive gastric cancer,the miR-191/PLCd1 ratio in cancer tissues of patients with infiltration depth T_(3)-T_(4)was significantly higher than that of patients with T_(1)-T_(2)(P<0.05).Additionally,the miR-191/PLCd1 ratio in patients with lymph node metastasis was significantly higher than that in patients without lymph node metastasis(P<0.05).Spearman rank correlation analysis revealed that the miR-191/PLCd1 ratio in cancer tissues was positively correlated with infiltration depth and lymph node metastasis(r=0.775,0.728,P<0.05).[WTHZ]Conclusion The miR-191/PLCd1 ratio in cancer tissues is closely associated with the biological behaviors of H.pylori-positive gastric cancer,and may serve as a novel target for diagnosis and treatment strategies in gastric cancer.
