彩虹桉组织培养试验

摘要 于2023年在福建省林业科技试验中心,以彩虹桉当年生、半木质化带芽点的茎段为外植体,开展组织培养技术体系研究,筛选适宜外植体灭菌、不定芽诱导、继代增殖、生根诱导等阶段的最优条件。结果表明:75%酒精消毒8 s后,用0.1%升汞消毒8 min可获得较多的无菌外植体,为最佳灭菌处理方法;彩虹桉在改良MS+6-BA 0.2 mg·L^(-1)+NAA 0.1 mg·L^(-1)+蔗糖30 g·L^(-1)中培养20 d后诱导率可达86.0%,为最佳诱导培养基;在改良MS+6-BA 0.25 mg·L^(-1)+IBA 0.05 mg·L^(-1)+NAA 0.10 mg·L^(-1)+蔗糖30 g·L^(-1)中培养20 d后,平均增殖系数达2.23,为最佳继代培养基;在改良MS+IBA 0.30 mg·L^(-1)+NAA 0.05 mg·L^(-1)+蔗糖20 g·L^(-1)+活性炭0.1 g·L^(-1)中培养30 d后,平均生根率达94.0%,根数3~6条,根长1~2 cm,为最佳生根培养。研究结果可为建立彩虹桉组织培养技术体系及规模化育苗提供技术支持。 In 2023,a tissue culture technology system study was conducted at the Fujian Forestry Science and Technology Test Center using stem segments of Eucalyptus deglupta with semiwoody bud points as explants.The optimal conditions for sterilization of explants,induction of adventitious buds,subculture proliferation,and rooting induction were screened.The results showed that after disinfecting with 75%alcohol for 8 seconds,disinfecting with 0.1%mercuric chloride for 8 minutes can obtain more sterile explants,which is the best sterilization treatment method;After 20 days of cultivation in improved MS+6-BA 0.2 mg·L^(-1)+NAA 0.1 mg·L^(-1)+sucrose 30 g·L^(-1),the induction rate of Eucalyptus deglupta reached 86.0%,making it the optimal induction medium;after 20 days of cultivation in an improved MS+6-BA 0.25 mg·L^(-1)+IBA 0.05 mg·L^(-1)+NAA 0.10 mg·L^(-1)+sucrose 30 g·L^(-1)medium,the average proliferation coefficient reached 2.23,making it the optimal subculture medium;After 30 days of cultivation in improved MS+IBA 0.30 mg·L^(-1)+NAA 0.05 mg·L^(-1)+sucrose 20 g·L^(-1)+activated carbon 0.1 g·L^(-1),the average rooting rate reached 94.0%,with 3~6 roots and 1~2 cm root length,making it the optimal rooting culture.The research results can provide technical support for establishing a tissue culture technology system for Eucalyptus deglupta and for large-scale seedling cultivation.
