PGC1α通过抑制膀胱癌细胞侵袭抑制膀胱癌的进展

摘要 目的探究过氧化物酶体增殖物激活受体γ共激活剂1α(PGC1α)在膀胱癌中的表达模式及其对膀胱癌细胞侵袭性的影响。方法通过对比分析膀胱癌患者和对照组人群(非膀胱癌患者)血清中PGC1α基因的表达水平差异及PGC1α的表达水平与膀胱癌临床分期和病理分级之间的关系;利用膀胱癌细胞系T24进行PGC1α的过表达实验,分析其对T24细胞增殖、克隆形成及侵袭能力的影响,并通过Western blot实验检测上皮-间质转化(EMT)相关蛋白表达。结果膀胱癌患者血清中PGC1α的表达水平显著低于对照组人群,且高表达PGC1α的患者病理分期和病理分级较低;表达PGC1α后,T24细胞的增殖、克隆形成和侵袭能力显著减弱。此外,过表达PGC1α导致与EMT相关的蛋白SNAIL1、N-cadherin和Vimentin的表达下调,E-cadherin的表达上调。结论(1)PGC1α表达水平降低是膀胱癌发生的重要因素,并且与更高的临床分期和病理分级有关;(2)PGC1α可以通过调节EMT相关蛋白抑制肿瘤细胞的增殖和侵袭。 Objective To investigate the expression pattern of peroxisome proliferator-activated receptor gamma coactivator-1 alpha(PGC1α)in bladder cancer and its impact on the invasiveness of bladder cancer cells.Methods The differential expression of the PGC1αgene in serum samples from bladder cancer patients and control subjects(non-bladder cancer patients)was analyzed,and the relationship between the expression level of PGC1αand the clinical stage and pathological grade of bladder cancer was examined.PGC1αoverexpression experiments were conducted in the bladder cancer cell line T24 to analyze its effects on cell proliferation,colony formation,and invasion.Western blot experiments were performed to detect the expression of Epithelial-Mesenchymal Transition(EMT)-related proteins.Results The expression level of PGC1αin serum samples from bladder cancer patients was significantly lower than that in the control group,and bladder cancer patients with high PGC1αexpression had lower clinical stages and pathological grades.Overexpression of PGC1αresulted in significantly decreased proliferation,colony formation,and invasion abilities of T24 cells.Furthermore,PGC1αoverexpression led to downregulation of EMT-related proteins SNAIL1,N-cadherin,and Vimentin,as well as upregulation of E-cadherin expression.Conclusion(1)Decreased expression of PGC1αis an important factor in the development of bladder cancer and is associated with higher clinical stages and pathological grades.(2)PGC1αcan inhibit tumor cell proliferation and invasion by regulating EMT-related proteins.
