摘要
目的探究溴结构域蛋白4(Bromodomain-containing protein 4,BRD4)在宫颈癌中的表达及其抑制剂JQ1对细胞增殖和迁移的影响。方法收集51例宫颈癌患者及51例子宫良性病变患者的宫颈组织标本,应用免疫组织化学染色方法检测正常宫颈组织及宫颈癌组织BRD4的表达情况,并应用统计学方法分析BRD4表达与宫颈癌患者的FIGO分期、浸润间质深度、分化程度、盆腔淋巴结转移、淋巴脉管间隙浸润是否具有相关性;体外培养宫颈癌Hela和SiHa细胞,CCK-8实验探究JQ1对癌细胞增殖能力的影响,划痕实验探究JQ1对癌细胞迁移能力的影响。结果与正常宫颈组织相比,BRD4在宫颈癌组织中的表达明显增高(P<0.001),但与FIGO分期、盆腔淋巴结转移、浸润间质深度、淋巴脉管间隙浸润、分化程度均无相关性(P>0.05);JQ1对Hela和SiHa细胞增殖具有抑制作用(P<0.05);JQ1对Hela和SiHa细胞迁移也具有抑制作用(P<0.05)。结论BRD4在宫颈癌组织中高表达,其抑制剂JQ1能抑制细胞增殖和迁移能力。
Objective Exploring the expression of BRD4 in cervical cancer and the effect of its inhibitor JQ1 on cell proliferation and migration.Methods Histological specimens were collected from 51 cases of cervical cancer and 51 cases of uterine benign lesions.The expression of BRD4 protein in normal cervical tissue and cervical cancer tissue was detected by immunohistochemical staining.Statistical methods were used to analyze the indexes.Analyze whether there is a correlation between BRD4 expression and FIGO stage,the depth of cervical stromal invasion,the degree of differentiation,lymph vascular space invasion and lymph node metastasis in cervical cancer patients;The cervical cancer cells(Hela and SiHa cell lines)were cultured in vitro.CCK-8 assay was used to detect the cell viability of Hela and SiHa cells after treated with different concentrations of JQ1.Wound-healing assay was used to detect the effect on the migration of Hela and SiHa cells after treated with JQ1.Results The expression of BRD4 was significantly higher in cervical cancer tissues compared to normal cervical tissues(P<0.001).BRD4 expression was not correlated with FIGO stage,pelvic lymph node metastasis,the depth of cervical stromal invasion,lymph vascular space invasion and the degree of differentiation(P>0.05).Hela and SiHa cells were treated with JQ1.The cell proliferation of Hela and SiHa cells was inhibited by JQ1(P<0.05).The migration of Hela and SiHa cells was also inhibited by JQ1(P<0.05).Conclusion BRD4 is expressed at high levels in cervical cancer tissues.The inhibitor,JQ1,has been demonstrated to inhibit both cell proliferation and migration.
