脂质纳米颗粒递送mRNA药物

Lipid nanoparticles for mRNA drugs delivery

摘要 在新冠病毒mRNA疫苗获得成功的同时,mRNA在各种疾病的预防和治疗中受到越来越多的关注。脂质纳米颗粒(lipid nanoparticles,LNPs)是目前最常用的mRNA药物递送方式之一。本文将简要介绍含LNPs在内的mRNA药物递送平台,总结LNPs的历史发展,探讨LNPs优化策略以及靶向器官或细胞类型的特异性等问题,最后聚焦mRNA-LNPs药物的主要应用。 Due to the success of mRNA vaccines against COVID-19 viruses,mRNA drugs have received great attention in the prevention and treatment of various diseases.Lipid nanoparticles(LNPs)are one of the most used strategies for the delivery of mRNA drugs to date.Here,we briefly overview some kinds of mRNA drugs delivery platforms including LNPs and summarize timeline of some crucial milestones for LNPs development.We discuss the optimization strategies of LNPs,as well as targeting specific tissues and cell types of LNPs delivery.We also highlight the key applications of mRNA-LNPs in clinical studies.
