可注射性钼/生物矿化硅/壳聚糖骨充填材料的制备及性能评价

摘要 目的:构建一种缓释钼(Mo)离子的可注射性骨充填材料并对其性能进行评价。方法:将生物矿化硅(DBs)负载不同比率的Mo离子,再与壳聚糖(CS)基水凝胶共混,构建Mo/DBs/CS缓释体系。通过扫描电镜(SEM)、能谱仪(EDS)、傅里叶红外光谱仪(FTIR)、电感耦合等离子体质谱仪(ICP)等仪器检测其理化性质,并通过细胞毒性实验及碱性磷酸酶(ALP)分泌水平、ALP和茜素红染色方法观察其生物相容性和促进大鼠骨髓间充质干细胞(BMSCs)成骨分化能力。结果:SEM、EDS、FTIR检测结果证明DBs可以成功负载Mo离子;DBs对Mo离子的包封率为8%左右。第21天时,Mo/DBs/CS对Mo离子累计释放率为81%,缓释效果最佳。Mo离子低于200 mg/L、DBs浸提液低于2500 mg/L时对BMSCs无细胞毒性(P<0.001),而且Mo离子浓度在25~3.13 mg/L可以提高BMSCs分泌ALP的水平(P<0.05)。ALP及茜素红染色结果表明Mo/DBs/CS缓释体系可以显著促进MSCs成骨分化。结论:Mo/DBs/CS缓释体系是一种具有良好生物相容性、骨诱导活性及可注射性等优点的骨缺损充填材料。 AIM:Construction and evaluation of an injectable slow-release molybdenum(Mo)bone filling material.METHODS:Diatom biosilica(DBs)was loaded with different ratios of Mo ions,then blended with chitosan(CS)-based hydrogel to build the Mo/DBs/CS slow-release system.The physical and chemical properties were examined by the scanning electron microscope(SEM),the energy spectrometer(EDS),the fourier infrared spectrum device(FTIR)and the inductively coupled plasma mass spectrometer(ICP).The biological compatibility and the ability of promoting bone differentiation of rat bone marrow stem cells were observed by cytotoxic experiment,the level of alkaline phosphatase(ALP)secretion,the ALP and Alizarin red staining.RESULTS:SEM,EDS and FTIR test results showed that Mo ions can be successfully loaded in DBs.The encapsulation rate of Mo by DBs was around 8%.At 21d,the total release rate of the Mo ion in Mo/DBs/CS group was 81%,which had the best slow release effect.Mo ion under 200 mg/L and the DBs leaching liquid under 2500 mg/L were not cytotoxic to BMSCs.Mo ion concentration at 25~3.13 mg/L could increase the secreting of ALP by BMSCs(P<0.001).ALP and Alizarin red staining results showed that Mo/DBs/CS system significantly promote the osteogenesis differentiation of BMSCs.CONCLUSION:The Mo/DBs/CS slow-release system is a kind of bone defect filling material with good biological compatibility and osteoinductive activity.
