黑果枸杞多糖的提取及抗肿瘤活性分析

摘要 为了研究黑果枸杞多糖的最佳提取工艺条件和枸杞多糖抗肿瘤活性,通过改变料液比、提取温度、提取时间等影响因素设计正交试验.提取的黑果枸杞多糖溶液经醇沉、抽滤、干燥等处理得粗多糖粉末,该粉末经大孔树脂脱色、Sevage法脱蛋白,精制得到黑果枸杞多糖.结果表明,黑果枸杞多糖的最佳提取工艺条件为:料液比1∶25,提取温度80℃,提取时间3 h,多糖提取率为11.58%;黑果枸杞多糖体外有抑制人慢性粒细胞白血病细胞株K562增殖的作用,有望成为一种理想的抗肿瘤佐剂. To study the optimal extraction process conditions and anti-tumor activity of polysaccharides from lycium ruthenicum,orthogonal experiments were designed by changing factors such as material liquid ratio,extraction temperature,and extraction time.The extracted lycium ruthenicum polysaccharide solution was treated with alcohol precipitation,filtration,and drying to obtain crude polysaccharide powder.The powder was decolorized by macroporous resin,deproteinized by Sevage method,and refined to obtain lycium ruthenicum polysaccharide.The results showed that the optimal extraction process conditions for polysaccharides from lycium ruthenicum were material liquid ratio 1∶25,extraction temperature 80 ℃,extraction time 3 hours,and the polysaccharide extraction rate was 11.58%;Lycium ruthenicum polysaccharides have an inhibitory effect on the proliferation of human chronic myeloid leukemia cell line K562 in vitro,and are expected to become an ideal anti-tumor adjuvant.
