UPLC-MS法分析发酵不同时间的人参果中12种人参皂苷含量

Analysis of 12 Ginsenosides in Ginseng Berry Fermented at Different Time by UPLC-MS

摘要 本研究建立同时分析12种人参皂苷的超高效液相-三重四极杆串联质谱(UPLC-MS)分析方法,比较人参果自然放置发酵前后的12种人参皂苷含量变化。将样品经醇回流提取,采用Thermo Syncronis-C18色谱柱(100 mm×2.1 mm,1.7μm),0.1%甲酸-水溶液(A)-乙腈(B)为流动相,采用梯度洗脱,流速0.2 mL/min;柱温30℃;进样量5μL;电喷雾离子源(ESI);喷雾电压2.5 kV;负离子扫描模式;扫描范围m/z 100.0~1500.0;采用Xcalibur分析软件处理数据。结果表明,人参皂苷的线性关系良好(R^(2)≥0.995),平均回收率在91.23%~109.13%,稳定性RSD值为0.95%~2.67%,重复性RSD值为1.37%~2.67%。人参果中人参皂苷含量随着放置发酵时间的不同,含量会发生变化,此分析方法可用于人参果中的人参皂苷含量测定,对于人参果质量控制具有指导作用。 This study established an ultra performance liquid chromatography triple quadrupole tandem mass spectrometry(UPLC-MS)method for simultaneous analysis of 12 ginsenosides,and to compare the content changes of 12 ginsenosides in ginseng berry before and after natural fermentation.The samplewas extractedbyalcohol reflux,and a ThermoSynchronis-C18chromatographic column(100mm×2.1mm,1.7μm)using 0.1%formic acid aqueous solution(A)and acetonitrile(B)as the mobile phase and gradient elution at a flowrate of 0.2 mL/min;column temperature:30℃;injection volume 5 L;electric spray ion source(ESI);spray voltage 2.5 kV;negative ion scanning mode;scanning range:m/z 100.0~1500.0;the data were processed using Xcalibur analysis software.The linear relationship among ginsenosides was good(R^(2)≥0.995),with an average recovery of 91.23%to 109.13%,a stability RSD of 0.95%to 2.67%,and a repeatability RSD of 1.37%to 2.67%.The contents of ginsenosides in Ginseng berry changed with the time of storage and fermentation.This analytical method could be used to determine the content of ginsenosides in ginseng berry,and has a guiding role in the quality control of ginseng berry.
