摘要
采用Agilent ZROBAX SB-Aq(4.6 mm×250 mm, 5μm)色谱柱;以乙腈-0.1%磷酸溶液为流动相,梯度洗脱;流速为0.8 mL/min,进样量为10μL,检测波长为350 nm,柱温为35℃。建立一测多评法同时测定景天三七中槲皮苷、杨梅苷、异槲皮苷、番石榴苷4个黄酮类成分含量。上述4个成分在相应的浓度范围内线性关系良好(R^(2)=1),以槲皮苷为内参物,杨梅苷、异槲皮苷、番石榴苷的相对校正因子分别为1.091、0.897及0.983,RSD分别为0.48%、0.28%、0.08%;28批不同产地的景天三七中4个有效成分采用一测多评法与外标法的实测值之间无显著性差异。综上,一测多评法测定景天三七中杨梅苷、异槲皮苷、番石榴苷、槲皮苷4个成分的含量准确可行,可用于景天三七的质量控制。
A“quantitative analysis of multicomponents by single-marker”(QAMS)method for simultaneous determination of myricitrin,isoquercitrin,guaijaverin,and quercitrin in Sedum aizoon L.was established,using Agilent ZROBAX SB-Aq(4.6 mm×250 mm,5μm)Chromatographic column.Using acetonitrile 0.1%phosphoric acid solution as the mobile phase,gradient elution,the flow rate was 0.8 mL/min,and the injection volume was 10μL,the detection wavelength was 350 nm and the column temperature was 35℃.The above four components showed good linear relationships within the corresponding concentration range(R^(2)=1),with quercitrin as the internal reference.The relative correction factors for myricitrin,isoquercitrin and guaijaverin were 1.091,0.897 and 0.983,respectively,with RSD of 0.48%,0.28%and 0.08%.There was no significant difference between the measured values of four active ingredients in 28 batches of Sedum aizoon L.from different origins and batches using the one test multiple evaluation method and the external standard method.The quantitative analysis of multicomponents by single-marker method was accurate and feasible for determining the content of four components in Sedum aizoon L.,including myricitrin,isoquercitrin,guaijaverin and quercitrin.It can be used for quality control of Sedum aizoon L..
